Abstract Ticks transmit more pathogens to humans and animals than any other arthropod. We describe the 2.1 Gbp nuclear genome of tick, Ixodes scapularis (Say), which vectors that cause Lyme disease, human granulocytic anaplasmosis, babesiosis diseases. The large reflects accumulation repetitive DNA, new lineages retro-transposons, gene architecture patterns resembling ancient metazoans rather pancrustaceans. Annotation scaffolds representing ∼57% genome, reveals 20,486 protein-coding genes expansions families associated with tick–host interactions. report insights from analyses into parasitic processes unique ticks, including host ‘questing’, prolonged feeding, cuticle synthesis, blood meal concentration, novel methods haemoglobin digestion, haem detoxification, vitellogenesis off-host survival. identify proteins agent an emerging encephalitis-causing Langat virus, a population structure correlated life-history traits transmission disease agent.

Significance Peptides and their specific receptors form molecular peptidergic systems (PSs) that are essential components of neuronal communication in the animal brain. Many PSs have been characterized insects mammals but precise evolutionary relationship is not fully understood. We interrogated genomic sequence databases used phylogenetic reconstruction tools to show a large fraction human were already present last common ancestor flies, mollusks, urchins, mammals. Neuropeptides and protein hormones are ancient molecules that mediate cell-to-cell communication. The whole genome sequence from the red flour beetle Tribolium castaneum , along with those other insect species, provides an opportunity to study evolution of genes encoding neuropeptide hormones. We identified 41 these in by using a combination bioinformatic peptidomic approaches. These encode >80 mature neuropeptides hormones, 49 peptides which were experimentally peptidomics central nervous system neuroendocrine organs. Twenty-three have orthologs Drosophila melanogaster : Sixteen five different groups likely result recent gene expansions during evolution. contain related antidiuretic factor-b (ADF-b), CRF-like diuretic hormone (DH37 DH47 ), adipokinetic (AKH), eclosion hormone, insulin-like peptide.
