Sheep are an important livestock species whose gastrointestinal tract is essential for overall health. Feed contaminants such as bacterial toxins and mycotoxins severely damage the sheep intestine, yet mechanisms remain mostly elusive partially due to lack of physiologically relevant in vitro models. Here, we investigated molecular underlying deoxynivalenol (DON)-induced toxicity by developing intestinal organoids from isolated crypts Hu sheep. The had a central lumen monolayer epithelium, could be continuously passaged, cryopreserved, resuscitated. Histological transcriptomic analysis showed that recapitulate cell lineages gene expression characteristics original tissues. Statistical indicated DON exposure significantly inhibited organoid formation efficiency, well proliferation activity cells. RNA-seq Western blotting further revealed induces inhibiting PI3K/AKT/GSK3β/β-catenin signaling pathway. Aflatoxin B1 (AFB1) and Ochratoxin A (OTA) are two of the most potent mycotoxins, recognized for their severe toxicity. In recent years, consumption bioactive substances has proven to be a valuable ally in combating harmful effects on human health. For this purpose, study evaluates protective fermented whey (FW) pumpkin (P), as functional ingredients bread, duodenum tissue against sub-chronic toxicity induced by AFB1 OTA. Nine groups male female Wistar rats (n = 5 per sex/group) were exposed different combinations AFB1, OTA, FW, P 28 days. The gene expression apoptotic antioxidant markers, including p53, Bax, Hmox1, NF-κB, occludin, was measured quantitative real-time PCR (RT-qPCR). + OTA exposure led an increased p53 with downregulation Bax Hmox1. Occludin expression, which supports tight junction integrity, remained largely unaffected. Zearalenone (ZEN) contamination in dried distiller’s grains and solubles (DDGS) poses serious health risks economic losses animal farming. This study aimed to evaluate the effectiveness of probiotic fermentation using Lactobacillus plantarum CN1 detoxifying ZEN optimizing conditions for maximum efficiency. L. CN1, identified with 99% genetic homology, was used DDGS fermentation. The detoxification mechanism analyzed through adsorption assays, post-heat treatment effects, scanning electron microscopy (SEM). removal assessed over 72 h under various conditions, including bacterial concentration, temperature, pH optimization. results showed that achieved a rate 69% within h, an optimized efficiency 75.6% at 4 × 10 9 CFU/mL. Over 60% adsorbed by cell wall, while supernatant intracellular fluid remained below 5%. Scanning (SEM) analysis highlighted structural changes cells, particularly elongation thinning, more pronounced damage observed following heat treatment. These modifications may explain varying efficiencies observed. Heat treatment, autoclaving, significantly enhanced 82.9%, whereas acid alkali treatments reduced it. Fermentation also improved nutritional quality DDGS, increasing crude protein 7.16%, reducing fiber 0.65%, lowering 4.3. findings demonstrate offers promising, cost-effective strategy mitigating enhancing quality. Fumonisin B1 (FB1) is a toxic metabolite produced by fungi of the Fusarium genus. FB1 common contaminant grains and their derived feed products, posing global concerns for environmental pollution food safety. Curcumin (Cur), natural polyphenolic compound, has anti-inflammatory, antioxidant, free radical scavenging properties. Although studies have explored apoptotic mechanisms FB1, underlying mitigation FB1-induced toxicity Cur are not fully understood, limiting its potential application as novel additive to prevent toxicity. In this study, porcine kidney cells (PK-15) were used experimental model, utilizing mRNA miRNA transcriptome technologies. The results revealed that upregulated miR-1249 inhibits target gene Ern1 in PK-15 cells, thereby suppressing IRE1/MKK7/JNK/CASPASE3 endoplasmic reticulum (ER) stress pathway alleviating cell apoptosis.
