ABSTRACT Environmental DNA (eDNA) analyses provide valuable ecological data. Recent studies have explored eDNA dynamics related to reproductive behavior and developmental stages, revealing significant variations in concentrations across different life stages. However, there is a gap understanding the association between changes before after events, such as egg hatching. This study addresses this by monitoring signals chum salmon ( Oncorhynchus keta ) during their early stages examining effects of physiological behavioral changes. For purpose, flux was monitored rearing experiments with (egg, alevin, fry). The varied significantly stages: while no detected stage, increased rapidly After hatching, became stable alevin stage but approximately 30‐fold when they progressed into fry (LMM post hoc Tukey‐HSD test: p < 0.05). These results suggest that vary can be utilized estimate monitor fish development even under natural conditions, those occurring gravel for salmonid species.

ABSTRACT Environmental DNA (eDNA) has evolved into a valuable asset of the ecologists' toolkit, enabling time‐ and cost‐efficient biodiversity assessments in wide variety ecosystems. Since eDNA can be isolated from broad range environmental substrates, its persistence times those media are decisive importance for drawing inferences about species presence. For first time, we characterize water sediment samples seminatural waterholes savanna system South Africa to gain better understanding decay these waterbodies. Using mesocosm experiments, tracked two different states (extracellular membrane bound), during seasons (wet dry), substrates (surface sediment). Extracellular degraded rapidly first‐order exponential fashion membrane‐bound exhibited slower decline with more intricate patterns, involving initial reduction followed by subsequent increase measured concentrations. Over the past century, numerous studies have proposed various organisms for biomonitoring of aquatic systems, but only recently has zooplankton emerged as a promising indicator water quality. The traditional identification methods, however, can be inefficient in context monitoring efforts, they are often time consuming and costly. DNA metabarcoding offers powerful alternative, providing more efficient reliable approach to monitor communities. In this review, we assess current state-of-the-art methodologies used evaluate marine brackish communities through workflow. While several emerging approaches been reported, no standardization achieved so far. extraction step gained most consensus, with widespread use commercial kits (DNeasy Blood & Tissue kit employed ca. 25% studies), though there is still significant variation selection. Additionally, 18S COI were main molecular markers (ca. 61% 54%, respectively) target region varied former. Moreover, many methodologies, particularly those processing samples, lack practical validation. Some also fail provide sufficient detail their methodology descriptions hindering reproducibility. Overall, shows great potential communities, further effort needed establish standardized practices optimize across entire methodological pipeline.
