Abstract Objectives To assess the long-term humoral immunity induced by booster administration, as well ability of binding antibody and surrogate virus neutralization tests (sVNT) to predict neutralizing antibodies (NAbs) against SARS-CoV-2 Omicron variant. Methods A total 269 sera samples were analyzed from 64 healthcare workers who had received a homologous dose BNT162b2. Neutralizing assessed sVNT anti-RBD IgG measured with sCOVG assay (Siemens Healthineers ® ) at five timepoints; before up 6 months following booster. Antibody titers correlated BA.1 variant obtained pseudovirus test (pVNT) reference method. Results While Wild-type percentage inhibition (POI) remained above 98.6% throughout follow-up period after NAbs pVNT showed respectively 3.4-fold 13.3-fold decrease compared peak reached day 14. followed steady decline until reaching POI 53.4%. Anti-RBD assays strongly (r=0.90) performed similarly presence (area under ROC: 0.82 for both assays). In addition, new adapted cut-off values (>1,276 BAU/mL) (POI>46.6%) found be better predictors activity. Conclusions This study significant drop in administration. highly could activity moderate performance.

Since the initiation of COVID-19 pandemic, there has been a need for development diagnostic methods to determine factors implicated in mounting an immune response against virus. The most promising indicator suggested be neutralizing antibodies (nAbs), which mainly block interaction between Spike protein (S) SARS-CoV-2 and host entry receptor ACE2. In this study, we aimed develop optimize conditions competitive ELISA measure serum titer, using recombinant trimeric modified have six additional proline residues (S(6P)-HexaPro) h-ACE2. results our surrogate Virus Neutralizing Assay (sVNA) were compared commercial sVNT (cPass, Nanjing GenScript Biotech Co., City, China), serially diluted sera from vaccinees, high correlation ID

Abstract Since early 2020, the world has been embroiled in an ongoing viral pandemic with SARS‐CoV‐2 and emerging variants resulting mass morbidity estimated 6 million deaths globally. The scientific community pivoted rapidly, providing unique innovative means to identify infected individuals, technologies evaluate immune responses infection vaccination, new therapeutic strategies treat individuals. Never before immunology so critically at forefront of combatting a global pandemic. It now become evident that not just antibody responses, but formation durability memory cells following vaccination are associated protection against severe disease from infection. Furthermore, emergence concern (VoC) highlight need for immunological markers quantify protective capacity Wuhan‐based vaccines. Thus, harnessing modulating response is key successful treatment disease. We here review latest knowledge about generation natural provide insights into attributes may protect variants.

Obesity is a known risk factor for severe respiratory tract infections. In this prospective study, we assessed the impact of being obese or overweight on longitudinal SARS-CoV-2 humoral and cellular responses up to 18 months after infection. 274 patients provided blood samples at regular time intervals including (BMI ≥30, n=32), 25-29.9, n=103) normal body weight 18.5-24.9, n=134) patients. We determined spike-specific IgG, IgA, IgM levels by ELISA neutralising antibody titres neutralisation assay. RBD- memory B cells were investigated ELISpot, spike- non-spike-specific IFN-γ, IL-2 IFN-γ/IL-2 secreting T FluoroSpot cell receptor (TCR) sequencing was performed. Higher BMI correlated with increased COVID-19 severity. Humoral stronger in than associated higher IgG binding relative titres. Linear regression models demonstrated that BMI, age severity independently immune responses. found an proportion unique specific clonotypes infection vaccination boosted irrespective although boosting observed Overall, our results highlight more disease over-reactivity system infection, underscoring importance recognizing overweight/obese individuals as group prioritisation vaccination.

The Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) virus has been causing the COVID-19 pandemic since December 2019, with over 600 million infected persons worldwide and six deaths. We investigated anti-viral effects of polyphenolic green tea ingredients synthetic resveratrol analogue 3,3',4,4',5,5'-hexahydroxy-trans-stilbene (HHS), a compound antioxidant, antitumor anti-HIV properties. In TCID50 assay, four out nine constituents showed minor to modest cell protective effects, whereas HHS demonstrated highest reduction (1103-fold) TCID50, indicating pronounced inhibition replication. was also highly effective inhibitor SARS-CoV-2 proliferation in VeroE6 cells an IC50 value 31.1 µM. HSS inhibited binding receptor-binding domain (RBD) spike protein human angiotensin-converting enzyme (ACE2) receptor (RBD-ACE2) 29% at 100 µM 9.2% 50 that inhibitory effect might least part be attributed ACE2. Based on chemical similarity other polyphenols, oral bioavailability is likely very low, resulting blood levels far below concentration EGCG against observed vitro. However, administration topically as nose or throat spray would increase concentrations several-fold above minimal (MIC) mucosa reduce load when administered soon after infection. Due these promising tissue culture results, further preclinical clinical studies are warranted develop additional treatment option for infection complement vaccines, which will main pillar combat pandemic.
