Melatonin, a pleiotropic neurohormone, is recognized for its hepatoprotective role in metabolic dysfunction-associated steatotic liver disease (MASLD) triggered by diet and/or chronodisruption. Nocturnin (Noct), circadian clock output regulating hepatic lipid metabolism, has an unclear MASLD. This study explores oscillations of Noct MASLD and synergy with melatonin. Differential gene expression analysis identified key lipogenic genes (HNF-4A SCD) as targets enzymatic activity. Male C57BL/6J mice were subjected to high-fat-high-fructose (H) or without chronodisruption (CD) 16 weeks. mRNA peaked at ZT12 Control H groups, while CD HCD groups showed peaks ZT6, alongside increased % relative amplitude, cumulative expression, shifted Centre Gravity (CoG). Melatonin treatment H, CD, significantly reduced amplitude expression. In OA-treated HepG2 cells, protein levels rose 36 h 32 h, respectively, CoG shifts. but correcting rhythmicity. Collectively, induces elevation phase shifts melatonin demonstrates corrective effects on phasing, underscoring therapeutic potential. .

Daylight is ubiquitous and crucial for mammalian vision as well non-visual input to the brain via intrinsically photosensitive retinal ganglion cells (ipRGCs) that express photopigment melanopsin. Meal timing emerges as a crucial factor influencing metabolic health that can be explained by the tight interaction between endogenous circadian clock and homeostasis. Mistimed food intake, such delayed or nighttime consumption, leads to desynchronization of internal is associated with an increased risk for obesity disturbances type 2 diabetes cardiovascular diseases. Conversely, meal aligned cellular rhythms optimize performance tissues organs. In this review, we provide overview effects discuss underlying mechanisms. Additionally, explore factors timing, including determinants chronotype genetics, well external influences like social factors, cultural aspects, work schedules. This review could contribute defining meal-timing-based recommendations public initiatives developing guidelines effective lifestyle modifications targeting prevention treatment Furthermore, it sheds light on must considered in design future intervention trials.

Today's challenge for precision medicine involves the integration of impact molecular clocks on drug pharmacokinetics, toxicity, and efficacy toward personalized chronotherapy. Meaningful improvements tolerability and/or medications through proper administration timing have been confirmed over past decade immunotherapy chemotherapy against cancer, as well commonly used pharmacological agents in cardiovascular, metabolic, inflammatory, neurological conditions. Experimental human studies recently revealed sexually dimorphic circadian responses. Dedicated randomized clinical trials should now aim to issue recommendations daily medical practice, integrating innovative technologies remote longitudinal monitoring metrics, statistical prediction clock function from single-timepoint biopsies, multiscale biorhythmic mathematical modelling. Importantly, chronofit patients with a robust function, who would benefit most chronotherapy, need be identified. Conversely, nonchronofit could emerging class chronobiotics targeting clock.

Abstract Dysfunction of circadian and sleep rhythms is an early feature many neurodegenerative diseases. Alzheimer's disease (AD) a progressive disorder resulting in cognitive psychiatric disturbances. Although it largely unclear whether dysfunctions contribute to the etiology AD or are consequence disease, there evidence that these conditions involved complex self‐reinforcing bidirectional relationship. According recent studies, dysregulation clock already occurs during asymptomatic stage could promote neurodegeneration. Thus, restoration preclinical may represent opportunity for intervention slow course.

Eating tightly interconnected recent evidence shows that meals crucial for control appetite regulation. Obesity results from combined long-term dysregulation intake (homeostatic hedonic circuits), energy expenditure, storage. Increasing supports loss synchrony significantly impairs homeostasis associated obesity. This review presents an overview mechanisms (homeostatic/hedonic) focuses on role response eating, thus providing fundamental research axis maintain healthy behavior.
