Allergen-specific immunotherapy represents the only method of achieving a lasting reduction in severity allergic symptoms. Abstract Bone marrow plasma cells (BMPCs) produce durable, protective IgM, IgG, and IgA antibodies, in some cases, pro-allergic IgE but their properties sources are unclear. We charted single BMPC transcriptional clonal heterogeneity food-allergic non-allergic individuals across CD19 protein expression given its inverse correlation to longevity. Transcriptional diversity revealed distinct functional profiles. Additionally, distribution of somatic hypermutation intraclonal antibody sequence variance suggest that CD19low CD19high BMPCs arise from recalled memory germinal center B cells, respectively. Most were peanut-allergic individuals; two out 32 independent donors bound peanut antigens vitro vivo. These findings shed light on origins highlight the bone as a source pathogenic allergy.

Background Peanut is an important source of dietary protein for human beings, but it also recognized as one the eight major food allergens. Sarcoplasmic calcium-binding protein (Cra a 4) from Crassostrea angulata belongs to the EF-hand superfamily, and understanding of its structure–allergenicity relationship is still insufficient. In this study, chemical denaturants were used destroy structure Cra 4, showing that disruption reduced IgG-/IgE-binding activity. The webinar series and workshop titled “Trust Your Gut: Establishing Confidence in Gastrointestinal Models – An Overview of the State Science Contexts Use” was co-organized by NICEATM, NIEHS, FDA, EPA, CPSC, DoD, Johns Hopkins Center for Alternatives to Animal Testing (CAAT) hosted at National Institutes Health Bethesda, MD, USA on October 11-12, 2023. New approach methods (NAMs) assessing issues gastrointestinal tract (GIT)- related toxicity offer promise addressing some limitations associated with animal-based assessments. GIT NAMs vary complexity, from two-dimensional monolayer cell line-based systems sophisticated 3-dimensional organoid derived human primary cells. Despite advances NAMs, challenges remain fully replicating complex interactions pro­cesses occurring within GIT. Presentations discussions addressed regulatory needs, challenges, innovations incorporating into risk assessment frameworks; explored state science using evaluating systemic toxicity, understanding absorption pharmacokinetics, potential allergenicity; discussed strengths, limitations, data gaps as well steps needed establish confidence these models use setting.
