Bifidobacteria support optimal infant vaccine responses

Accumulating
evidence
indicates
that
antibiotic
exposure
may
lead
to
impaired
vaccine
responses1-4;
however,
the
mechanisms
underlying
this
association
remain
poorly
understood.
Here
we
prospectively
followed
191
healthy,
vaginally
born,
term
infants
from
birth
15
months,
using
a
systems
vaccinology
approach
assess
effects
of
on
immune
responses
vaccination.
Exposure
direct
neonatal
but
not
intrapartum
antibiotics
was
associated
with
significantly
lower
antibody
titres
against
various
polysaccharides
in
13-valent
pneumococcal
conjugate
and
Haemophilus
influenzae
type
b
polyribosylribitol
phosphate
diphtheria
toxoid
antigens
combined
6-in-1
Infanrix
Hexa
at
7
months
age.
Blood
exposed
had
an
inflammatory
transcriptional
profile
before
vaccination;
addition,
faecal
metagenomics
showed
reduced
abundance
Bifidobacterium
species
these
time
vaccination,
which
correlated
6
later.
In
preclinical
models,
were
strongly
dependent
intact
microbiota
could
be
restored
germ-free
mice
by
administering
consortium
or
probiotic
already
widely
used
units.
Our
data
suggest
microbiota-targeted
interventions
mitigate
detrimental
early-life
immunogenicity.

The
outbreaks
of
respiratory
pathogens
like
severe
acute
syndrome
coronavirus
2
(SARS-CoV-2)
and
influenza
virus
(IV)
have
heightened
the
demand
for
highly
effective
vaccines
that
provide
strong
durable
immunity
in
human
populations.
However,
immune
responses
to
vaccination
vary
significantly
among
individuals
Recent
studies
demonstrated
gut
microbiota
play
an
essential
role
regulating
vaccination-induced
through
systemic
effects
gut-lung
axis
on
distant
organs,
lungs.
In
this
review,
we
first
synthesize
changes
composition
occur
during
pathogen
infections
vaccination.
Then,
discuss
underlying
immunological
mechanisms
bidirectional
immunomodulatory
between
vaccines.
Finally,
explore
strategies
designing
next-generation

pathway.
