This study aimed to investigate the protective effect of bergenin on myocardial ischemia/reperfusion (I/R) injury and elucidate its underlying mechanism. The in vivo model I/R was established by transient ligation left anterior descending coronary artery Sprague-Dawley rats, which were divided into sham, I/R, I/R+bergenin, I/R+bergenin+erastin (an agonist ferroptosis) groups.After established, rats underwent echocardiography assess cardiac function. Hematoxylin eosin (HE) staining Masson's trichrome performed evaluate pathological damage. Malondialdehyde (MDA), reactive oxygen species (ROS), glutathione (GSH) iron levels measured determine ferroptosis level. Western blotting used detect expression related proteins. Next, H9C2 cells subjected oxygen-glucose deprivation/reoxygenation (OGD/R) mimic vitro injury. EX527, a SIRT1 inhibitor, further explore role protection bergenin. In this part experiment, four groups: control, OGD/R, OGD/R+bergenin, OGD/R+bergenin+EX527. experiments, we found that group showed obvious damage, oxidative stress ferroptosis, while pretreatment reversed above injury, but inhibited inducer erastin. compared with OGD/R group, reduced level, mitochondrial dysfunction cells. We myocardium abrogated EX527. Moreover, activated SIRT1, increased phosphorylation AMPK level PGC-1α. Bergenin exerted modulating process during through SIRT1/AMPK/PGC-1α pathway.

Abstract Background: Limited research has been conducted on the interaction between ferroptosis and epithelial-mesenchymal transition (EMT) their combined effect esophageal squamous cell carcinoma (ESCC) patient prognosis. The present study aimed to develop a prognostic model based impact of EMT ESCC prognosis for clinical application. Methods: Gene expression levels data patients were obtained from GSE53625 dataset in gene omnibus (GEO) database, cancer genome atlas (TCGA) as validation set. By combining results cox regression analysis least absolute shrinkage selection operator (LASSO) analysis, we selected nine genes associated with prognosis, which then used construct model. Immune infiltration was evaluated using CIBERSORT single-sample Set Enrichment Analysis methods. Results: Nine key screened integrated score (FEIS). Compared low-FEIS group, high-FEIS group demonstrated shorter overall survival period. immune showed an increase elevated checkpoint molecules group. A nomogram constructed accurately predict Conclusion: Our introduced novel tool that integrates -and EMT-related biomarker, offered valuable insights developing personalized treatment strategies patients.
