Nuclear Factor Y (NF-Y) represents a group of transcription factors commonly present in higher eukaryotes, typically consisting three subunits: NF-YA, NF-YB, and NF-YC. They play crucial roles the embryonic development, photosynthesis, flowering, abiotic stress responses, other essential processes plants. To better understand genome-wide NF-Y domain-containing proteins, protein physicochemical properties, chromosomal localization, synteny, phylogenetic relationships, genomic structure, promoter cis-elements, interaction network NtNF-Ys tobacco (Nicotiana tabacum L.) were systematically analyzed. In this study, we identified 58 tobacco, respectively, divided into subfamilies corresponding to their relationships. Their tissue specificity expression pattern analyses for leaf drought saline-alkali stress, ABA response carried out using RNA-seq or qRT-PCR. Background/Objectives: Stay-green proteins (SGRs) play a vital role in regulating plant chlorophyll degradation and senescence. However, this gene family has not been explored Ipomoea species sweet potato. Methods: A total of 19 SGR genes were identified using Basic Local Alignment Search Tool (BLAST) methods. The proteins’ physiological properties, evolutionary phylogenetic relationships, conserved domain motifs, structures, collinearity, promoter cis-elements systematically analyzed. SUMMARY Drought is a major abiotic stress that impairs plant growth and development. Developing drought‐tolerant crop varieties an important goal of breeders. Transcription factors belonging to the DNA‐binding with one zinc finger (Dof) family regulate responses However, roles regulatory mechanisms Dof2.1 members in tolerance are still unclear. Here, we cloned IbDof2.1 gene from sweetpotato found its overexpression significantly enhanced drought sweetpotato, whereas ‐RNA interference (RNAi) plants displayed opposite phenotype. The ‐overexpression showed increased abscisic acid (ABA) proline contents stomatal sensitivity ABA decreased H 2 O accumulation. Furthermore, interacted ABA‐binding factor (IbABF2) promoted expression biosynthesis IbP5CS1 increase content, further activating reactive oxygen species (ROS) scavenging system. These results suggest IbDof2.1–IbABF2 module induces closure activates ROS system by regulating enhance sweetpotato. Our findings provide novel insights into plants.

Nuclear factor Y (NF-Y) is a heterotrimeric complex composed of three unique subunits: NF-YA, NF-YB, and NF-YC. This transcription binds to the CCAAT box eukaryotic promoters, playing crucial role in various biological processes plants. Despite its importance, NF-Y gene family has not been reported sweet potato (Ipomoea batatas) genome, an important food energy crop. Understanding function potatoes could provide valuable insights for genetic improvement yield enhancement. To address this gap, our research aimed comprehensively catalog characterize genes potatoes, which we refer as ‘IbNF-Y’, where ‘Ib’ denotes Ipomoea batatas. A total 37 NF-Ys were identified, including 11 21 5 NF-YC members, their phylogeny, structure, chromosomal distribution, conserved motifs analyzed. Additionally, assessed expression patterns under salt stress both light dark conditions using transcriptome sequencing. Notably, discovered that certain IbNF-Y showed significant changes stress, suggesting potential roles potato’s adaptation saline environments.
