Abstract Cellular mechanical forces play crucial roles in both normal physiology and disease, yet drug discovery efforts targeting mechanobiology have been limited part by assumptions about the conservation of contractile pathways across cell types. Here, we present first high-throughput contractility screen an annotated kinase inhibitor library, evaluating 2,418 compounds five primary human types using FLECS (Fluorescent Elastomer Contractility Sensors) platform. Quantification responses revealed selective divergent among related Clustering analysis identified distinct mechanobiological profiles novel pathway associations that challenge assumption are too highly conserved for targeting. This systematic approach supports wider adoption phenotypic screening as a viable strategy discovering cell-type specific modulators broad range mechanically-driven disease indications.

Introduction Patients with oral cancer often experience intense functional pain due to mechanical stimulation at the site. During aging, the cardiac extracellular matrix (ECM) undergoes gradual remodeling that reduces heart's ability to function. Specific ECM changes cause alterations in cellular signaling pathways, eliciting maladaptive responses. Here, we provide insight into current knowledge of how age-specific contribute altered ligand-receptor interactions, dysregulated mechanotransduction, and propagation pro-fibrotic cascades underpin dysfunction.
