Mapping the brain, one neuron at a time Spatial transcriptomics can link molecularly described cell types to their anatomical positions and functional roles. Moffitt et al. used combination of single-cell RNA-sequencing MERFISH (multiplexed error-robust fluorescence in situ hybridization) map identity location specific within mouse preoptic hypothalamus surrounding areas brain (see Perspective by Tasic Nicovich). They related these behaviors via gene activity. The approach provides an unbiased description area, which are important for sleep, thermoregulation, thirst, social behavior. Tremendous progress has been made since Neuron published our Primer on genetic dissection of neural circuits 10 years ago. Since then, cell-type-specific anatomical, neurophysiological, and perturbation studies have carried out in a multitude invertebrate vertebrate organisms, linking neurons to behavioral functions. New methods allow systematic classification cell types provide access diverse neuronal for connectivity coding during behavior. Here we evaluate key advances over the past decade discuss future directions.

Neuron activity across the brain How is it that groups of neurons dispersed through interact to generate complex behaviors? Three papers in this issue present brain-scale studies neuronal and dynamics (see Perspective by Huk Hart). Allen et al. found thirsty mice, there widespread neural related stimuli elicit licking drinking. Individual encoded task-specific responses, but every area contained with different types response. Optogenetic stimulation thirst-sensing one reinstated drinking previously signaled thirst. Gründemann investigated mouse basal amygdala relation behavior during tasks. Two ensembles showed orthogonal exploratory nonexploratory behaviors, possibly reflecting levels anxiety experienced these areas. Stringer analyzed spontaneous firing, finding primary visual cortex both information motor facial movements. The variability responses mainly arousal reflects encoding latent behavioral states. Social transmission of pain and relief In mice, both fear can be transferred by short social contact from one animal to a bystander. Neurons in brain region called the anterior cingulate cortex bystander mediate these transfers. However, specific projections involved such empathy-related behaviors are unknown. Smith et al. found that nucleus accumbens necessary for transfer mice (see Perspective Klein Gogolla). Fear, however, was mediated basolateral amygdala.
