How barriers to gene flow arise and are maintained key questions in evolutionary biology. Speciation research has mainly focused on that occur either before mating or after zygote formation. In comparison, postmating prezygotic (PMPZ) isolation-a barrier acts gamete release but formation-is less frequently investigated may hold a unique role generating biodiversity. Here we discuss the distinctive features of PMPZ isolation, including primary drivers molecular mechanisms underpinning isolation. We then present first comprehensive survey isolation research, revealing it is widespread form across eukaryotes. The also exposes obstacles studying part attributable challenges involved directly measuring uncovering its causal mechanisms. Finally, identify outstanding knowledge gaps provide recommendations for improving future This will allow us better understand nature this often-neglected reproductive contribution speciation.

Abstract Blotches in floral organs attract pollinators and promote pollination success. Tree peony (Paeonia suffruticosa Andr.) is an internationally renowned cut flower with extremely high ornamental economic value. Blotch formation on P. petals predominantly attributed to anthocyanin accumulation. However, the endogenous regulation of blotch remains elusive. Here, we identified regulatory modules governing anthocyanin-mediated petals, which involves transcription factors PsMYB308, PsMYBPA2, PsMYB21. PsMYBPA2 activated PsF3H expression provide sufficient precursor substrate for biosynthesis. PsMYB21 both PsFLS expressions promoted flavonol The significantly nonblotch regions inhibited by competing biosynthesis substrates, while conversely, its low region formation. In Lepidoptera (butterflies and moths), a genomic region around the gene cortex is hotspot locus, repeatedly used to generate intraspecific melanic wing color polymorphisms across 100-million-years of evolution. However, identity effector regulating within this locus remains unknown. Here, we show that none four candidate protein-coding genes including cortex, serve as major effectors. Instead, micro-RNA (miRNA), mir-193, serves three deeply diverged lineages butterflies, its function conserved in Drosophila. Lepidoptera, mir-193 derived from gigantic long non-coding RNA, ivory, it functions by directly repressing multiple pigmentation genes. We miRNA can drive repeated instances adaptive evolution animals.

Abstract Evolutionary variation in the wing pigmentation of butterflies and moths offers striking examples adaptation by crypsis mimicry. The cortex locus has been independently mapped as controlling colour polymorphisms 14 lepidopteran species, suggesting it acts a genomic hotspot for diversification patterns, but functional validation through protein-coding knockouts proven difficult to obtain.
