Abstract Plants are unique organisms that have developed ingenious strategies to cope with environmental challenges, such as herbivorous insects. One of these is the synthesis a vast array chemical compounds, known specialized metabolites, serve many ecological functions. Among most fascinating and diverse groups metabolites alkaloids, which characterized by presence nitrogen atom within heterocyclic ring. While some medicinal recreational applications, others highly unpalatable and/or toxic. The effects alkaloids on both humans insects can be very diverse, affecting their physiology behavior. Insects feed alkaloid-containing plants evolved various mechanisms consequences toxins. These include sequestration, where store in tissues or organs, enzymatic detoxification through enzymes cytochrome P450 monooxygenases glutathione S-transferases, behavioral adaptations selective feeding. In this review, we explore relationships between plant evolutionary enable exploit alkaloid-rich food sources niches minimizing harmful natural compounds. Synthetic biology has played an important role in the renaissance of natural products research during post-genomics era. The development and integration new tools have transformed workflow product discovery engineering, generating multidisciplinary interest field. In this review, we summarize recent developments biosynthesis from three different aspects. First, advances bioinformatics, experimental, analytical to identify associated with predicted biosynthetic gene clusters (BGCs) will be covered. Buckwheat is a promising crop with grains that are rich in nutrients and bioactive compounds. Genome sequence data for common Tartary buckwheat have recently become available. Currently, there critical need the development of simple reliable transient gene expression protocol, as well stable genetic transformation method, to facilitate metabolic engineering compounds, functional analysis genes, targeted editing, and, long-term perspective, accelerate breeding process buckwheat. The demand for novel enzyme-catalyzed reactions in chemical synthesis has spurred the development of many new-to-nature reactions. Additionally, detailed analysis biosynthetic pathways can uncover unprecedented chemical/enzymatic mechanisms. In this study, we revisited catalytic mechanism 2-oxoglutarate-dependent dioxygenase Pt2OGD-1, involved biosynthesis huperzine alkaloids. Our experimental and computational investigations uncovered a previously unknown enzymatic C-C bond cleavage piperidine ring alkaloid scaffold, resembling an oxidative retro-aza-Prins reaction. Here, transformation is initiated by hydrogen abstraction, followed electron transfer at 4-position heterocycle, triggering opening finally resulting loss carbon atom as formaldehyde. This discovery expands toolbox reactions, enhances our understanding these enzymes, may facilitate their application biotechnological production pharmaceutically relevant scaffolds well biocatalysts with similar activities.

Elucidating the biochemical pathways of specialised metabolites in plants is key to enable or improve their sustainable biotechnological production. Chemical tools can greatly facilitate discovery biosynthetic genes and enzymes. Here, we summarise transdisciplinary approaches where methods from chemistry chemical biology helped overcome challenges pathway elucidation. Based on recent examples, describe how state-of-the-art isotope labelling experiments guide selection gene candidates, affinity-based probes identification novel enzymes, semisynthesis availability elusive intermediates, biomimetic reactions provide a better understanding inherent reactivity. We anticipate that wider application such will accelerate pace elucidation plants.
