Demyelination is a common pathological feature in wide range of diseases, characterized by the loss myelin sheath and myelin-supporting oligodendrocytes. These losses lead to impaired axonal function, increased vulnerability axons damage, result significant brain atrophy neuro-axonal degeneration. Multiple pathomolecular processes contribute neuroinflammation, oligodendrocyte cell death, progressive neuronal dysfunction. In this study, we use cuprizone mouse model demyelination investigate long-term non-invasive gamma entrainment using sensory stimulation as potential therapeutic intervention for promoting myelination reducing neuroinflammation male mice. Here, show that multisensory mitigates demyelination, promotes oligodendrogenesis, preserves functional integrity synaptic plasticity, attenuates ferroptosis-induced reduces inflammation. Thus, protective effects on anti-neuroinflammatory properties support its approach demyelinating disorders.

Alzheimer's disease (AD) is the most common type of dementia and one leading causes death in elderly patients. The number patients with AD United States projected to double by 2060. Thus, understanding modifiable risk factors for an urgent public health priority. In parallel patients, cancer survivors estimated increase significantly, up 80% treated chemotherapy will develop cognitive deficits, termed chemotherapy-related impairment (CRCI). This Review discusses biologically plausible pathways underlying both disorders, goal why a proportion may be at higher developing AD. Highlighted are E4 allele apolipoprotein E gene, neuroinflammation, oxidative stress, DNA damage, mitochondrial dysfunction, neuronal synaptic loss, cellular senescence, brain-derived neurotrophic factor signaling, white matter blood-brain barrier/vascular tau pathology, transposable element reactivation.

Alzheimer’s disease (AD) is a progressive neurodegenerative disorder affecting millions worldwide. There no known cure for AD, highlighting an urgent need new, innovative treatments. Recent studies have shed light on promising, noninvasive approach using sensory stimulation as potential therapy AD. Exposing patients to and sound pulses at frequency of 40 hertz induces brain rhythms in the gamma range that are important healthy activity. Using this treatment animal models, we now beginning understand molecular, cellular, circuit-level changes underlie improvements pathology, cognition, behavior. A mechanistic understanding basic biology underlies 40-hertz will inform ongoing clinical trials offer promising avenue without side effects high costs typically associated with pharmacological interventions. Concurrent advancements neurotechnology can also noninvasively stimulate illuminating new possibilities alternative therapies. Altogether, these approaches could herald era treating making them beacon hope patients, families, caregivers facing challenges debilitating condition.

Gamma oscillations in brain activity (30–150 Hz) have been studied for over 80 years. Although the past three decades significant progress has made to try understand their functional role, a definitive answer regarding causal implication perception, cognition, and behavior still lies ahead of us. Here, we first review basic neural mechanisms that give rise gamma then focus on two main pillars exploration. The pillar examines major theories role information processing brain, also highlighting critical viewpoints. second reviews novel research direction proposes therapeutic oscillations, namely entrainment using sensory stimulation (GENUS). ABSTRACT Modulatory neurotransmitters exert powerful control over neurons and the brain vasculature. Gamma Entrainment Using Sensory Stimuli (GENUS) promotes amyloid clearance via increased perivascular cerebral spinal fluid (CSF) flux in mouse models of Alzheimer’s Disease. Here we use whole-brain activity mapping to identify cholinergic basal forebrain as a key region responding GENUS. In line with this, GENUS promoted cortical acetylcholine release, vascular dilation, vasomotion clearance. Inhibiting signaling abolished effects GENUS, including promotion arterial pulsatility, periarterial CSF influx, reduction levels. Our findings establish an essential component brain’s ability promote non-invasive sensory stimulation.
