C4-dicarboxylic acids are essential organic compounds characterized by a four-carbon structure and two carboxyl groups. Their export from cells is mediated specialized transporter proteins known as acid transporters (DCTs). The objective of this study was to investigate the specificity six DCTs (DCT1-5 C4t318) Aspergillus niger or oryzae, focusing on their role in different production strategies for acids. results indicate that compared WT strain, overexpressing dct2 dct3 A. CGMCC NO. 40550 specifically enhances succinic acid, increasing its yield 5.69 g/L 6.28 g/L, L-malic 11.02 12.11 g/L. Additionally, dct5 appears be involved transport both (6.19 g/L) (16.33 g/L). total yields T-D3-7, which lacks oxaloacetate hydrolase gene, were improved 27.75 25.19 T-D3-26, due blocking branch metabolism. Furthermore, heterologous expression oryzae C4T318 increased approximately 22.5%. best observed when strains T-D3-7 T-D5-16 scaled up 30 L bioreactor 84 h. reached 14.51 70.79 41.59 81.08 respectively. Moreover, purity produced 71%. This work further clarifies provides valuable insights optimizing production.
