Background:
Neoadjuvant
chemoimmunotherapy
(NACI)
is
promising
for
resectable
nonsmall
cell
lung
cancer
(NSCLC),
but
predictive
biomarkers
are
still
lacking.
The
authors
aimed
to
develop
a
model
based
on
pretreatment
parameters
predict
major
pathological
response
(MPR)
such
an
approach.
Methods:
enrolled
operable
NSCLC
treated
with
NACI
between
March
2020
and
May
2023
then
collected
baseline
clinical-pathology
data
routine
laboratory
examinations
before
treatment.
efficacy
safety
of
this
cohort
was
reported
variables

by
Logistic
Lasso

Dendritic
cells
(DCs)
are
professional
antigen-presenting

our
previous
work,
we
have
demonstrated
pDC
infiltration
promotes
glioblastoma
(GBM)
tumor
immunosuppression
through
decreased
IFN-α
secretion
via
TLR-9
signaling
increased
suppressive
function
regulatory
T
(Tregs)
IL-10
secretion,
resulting
in
poor
overall
outcomes
mouse
models
GBM.
Further
dissecting
the
mechanism
pDC-mediated
GBM
immunosuppression,
this
study,
identified
CCL21
as
highly
upregulated
by
multiple
cell
lines,
which
recruit
sites
CCL21-CCR7
signaling.
Furthermore,
activated
microenvironment
intracellular
β-arrestin
CIITA.
Finally,
found
CCL21-treated
directly
suppress
CD8+
proliferation
without
affecting
differentiation,
is
considered
canonical
pathway
immunotolerant
regulation.
Taken
together,
results
