Abstract The contrasting relationships of plant and animal protein intake with blood pressure (BP) may be partially attributed to the differential non-protein (e.g., saturated fat fibre) amino acid (AA) compositions. This study determined whether were related metabolomic profiles associated BP. included 1008 adults from African-PREDICT (aged 20–30 years). Protein was using 24-h dietary recalls. Twenty-four-hour ambulatory BP measured. Amino acids acylcarnitines analysed in spot urine samples liquid chromatography-tandem mass spectrometry-based metabolomics. Participants a low plant, high had higher SBP (by 3 mmHg, p = 0.011) than those (low-risk group). We found that mediated by BMI intake, which independently SBP. therefore not multiple regression analysis after adjusting for confounders. Abstract Background Cyclophosphamide (CTX) is the first-line medication for treatment of breast cancer, although it potentially leads to severe ovarian dysfunction and even premature failure (POF). However, mechanism CTX-induced POF remains unclear. Mesenchymal stem cell-based therapy has been wildly used treating numerous diseases. Therefore, our study aims elucidate underlying explore therapeutic effect human urine cells (hUSCs) in POF. Methods or granulosa cell (GCs) apoptosis were treated with hUSCs their exosomes vitro vivo. Morphological, histological, functional alternations examined using multiple approaches. The effector molecules hUSC-derived (hUSC-Exo) determined by differential expression analysis ovaries. target genes miRNA accessed transcriptome sequencing GCs, mechanisms further elucidated. Results remarkably inhibited promoted proliferation respectively. In addition, we observed that miR-27b-3p was highly expressed hUSC-Exo markedly suppressed GC specifically inhibiting SLC1A4, a serine transporter, which, turn, elevated concentration intracellular outflux cellular serine. More importantly, knockdown SLC1A4 simple supplementation GCs. Finally, demonstrated GCs essential reducing via elevating whereas protected against miR-27b-3p/SLC1A4/serine axis-mediated activation PI3K/AKT/mTOR signaling pathway. Conclusions Our suggests hUSC-based may provide an efficient approach prevention clinically. Graphical abstract

Increasing evidence suggests that frailty results from a complex age-associated metabolic decline. Here, we investigated the serum metabolomic profile of well-characterized cohort elderly subjects encompassing whole fit-to-frail continuum. Enrichment analyses revealed dysregulation amino acids and energy metabolism in both pre-frail frail participants. Remarkably, upregulated betaine levels emerged as specific biochemical signature females, holding promise for development novel targeted interventions.

Objective Ultra-high-performance liquid chromatography-time-of-flight mass spectrometry (UHPLC-TOF-MS) was employed to analyze serum metabolites and metabolic pathways associated with syndrome (MS) in the Yi Han populations of Yunnan. Methods Participants included individuals diagnosed MS healthy controls from Serum nontargeted amino acid-targeted metabolomics analyses were conducted identify differential (DEMs) pathogenesis these two ethnic groups. Results Nontargeted analysis revealed 2,762 DEMs group population, while 1,535 identified population [variable importance projection (VIP)>1, P <0.05]. Venn highlighted common unique between populations. KEGG pathway seven significantly enriched five group, primarily involving acid synthesis metabolism. To investigate role acids MS, levels 71 endogenous quantified. In 19 identified, related D-glutamine D-glutamate metabolism, as well cysteine methionine six significant enrichment sulfur valine, leucine, isoleucine biosynthesis. Conclusion Our study investigates differences through targeted approaches, identifying both especially disorders, including glycine, serine, threonine which may play critical roles regulating different dysfunctions worth further exploration pathogenesis, might provide insights for effective prevention treatment various
