Important health disparities are observed in the prevalence of obesity and associated non-communicable diseases (NCDs), including type 2 diabetes (T2D) metabolic dysfunction-associated steatotic liver disease (MASLD) among ethnic groups. Yet, underlying factors accounting for these remain poorly understood. Fructose has been widely proposed as a potential mediator NCDs, given that hepatic fructose catabolism can result deleterious effects, insulin resistance steatosis. Moreover, fermentation by gut microbiota produce metabolites such ethanol acetate, both which serve substrates de novo lipogenesis (DNL) could therefore contribute to development conditions. Significant inter-ethnic differences composition have observed. consumption varies across groups, intake demonstrated significantly alter composition, influence its fermenting properties effects. Therefore, may be influenced variations consumption, disparities. This review provides an overview complex interactions between host microbial catabolism, role ethnicity shaping processes their impact on health. Understanding provide insights into mechanisms driving improve personalized nutrition strategies. KEY POINTS: Dietary increased substantially over recent decades, with rising (NCDs) disease. Pronounced different groups NCD dietary underscore need elucidate Together well-known toxic effects emerging evidence highlights small intestinal sugars like various bacterial products There significant that, combined varying mediate To comprehensively understand mediating fructose-induced adverse future research should focus microbiota. Fructose has been considered as an additive from soda beverages. For the approval of new additives or to extend usage approved one, it is necessary conduct toxicological studies in order evaluate DNA damage induced by these compounds. Our study based on evaluating safety and nutraceutical potential (FRU), a cola beverage (Pepsi-cola, PEP), diet (Diet Coke, DCC), characterizing changes Drosophila melanogaster organism model human leukemia HL-60 cells performing different assays. results showed neither toxicity nor mutagenic activity FRU, PEP, DCC melanogaster, while only PEP exhibited protective effects antitoxity assay, showing 80% survival rate combined treatments. but not enhanced lifespan parameters up 23 more days at 5 mg/mL concentration. All three substances chemopreventive properties some checkpoints carried out related clastogenicity methylation patterns cells. In conclusion, tested compounds were safe concentrations moderate activity.

The study of type 2 diabetes mellitus (T2DM) pathophysiology relies mainly on the use animal models, most common which involves consumption high-fat diets comprising 60% calories from fat. Although these models reproduce onset and complications associated with T2DM, they do not accurately mimic human dietary patterns, as lack addition carbohydrates such fructose. This aimed to develop a C57BL/6 mouse model T2DM that mimics disease, occurs in younger individuals, via medium-fat diet (34.5% kcal fat) combined 20% fructose solution drinking water single low-dose streptozotocin (STZ) (100 mg/kg), diabetogenic drug. At week 20, D + T mice exhibited significant weight gain elevated fasting blood glucose levels compared those control development insulin resistance. Similarly, circulating hepatic enzymes (GPT, GOT, alkaline phosphatase), total cholesterol, LDL increased. Multi-organ damage, including reduced pancreatic islet size number, severe steatosis, inflammatory infiltration visceral adipose tissue, cardiac renal dysfunction, were also detected. proposed replicates young by combining STZ.
