Receptor-associated protein inhibits binding and uptake of all known endogenous ligands by megalin cubilin receptors via unknown mechanism(s). Here, we took advantage recently generated knockout cell lines to dissect the effect this on megalin- cubilin-mediated endocytosis. Our study reveals a novel role for receptor-associated in blocking megalin-stimulated endocytic fluid-phase markers receptor-bound proximal tubule cells addition its direct ligand receptors.

Albumin is derived from human or animal blood, and its ability to bind a large number of endogenous exogenous biomolecules makes it an ideal drug carrier. As result, albumin-based delivery systems are increasingly being studied. With these in mind, detailed studies the transport mechanism carriers particularly important. albumin receptors, glycoprotein 60 (GP60) secreted protein acidic rich cysteine (SPARC) play crucial role carriers. GP60 expressed on vascular endothelial cells enables cross cell layer, SPARC overexpressed many types tumor cells, while minimally normal tissue cells. Thus, this review supplements existing articles by detailing research history specific biological functions advances antitumor drugs using as Meanwhile, deficiencies future perspectives study interaction with also pointed out.

Cadmium (Cd) is a metal with no nutritional value or physiological role. However, it found in the body of most people because contaminant nearly all food types and readily absorbed. The burden Cd determined principally by its intestinal absorption rate as there mechanism for elimination. Most acquired accumulates within kidney tubular cells, where levels increase through to age 50 years but decline thereafter due release into urine injured cells die. This associated progressive disease, which signified sustained estimated glomerular filtration (eGFR) albuminuria. The intestinal absorption rate determines the body burden of Cd humans lack physiologic mechanism for excretion metal. Most acquired accumulates kidneys, which content increases through age 50 falls thereafter. decline from maximal kidney probably due to release injured tubular cells into urine and replacement destroyed nephrons by scar tissue. Chronic disease diagnosed when estimated glomerular filtration (eGFR) below 60 mL/min/1.73 m2, albuminuria present. Albuminuria defined as an albumin-to-creatinine ratio ≥ 30 mg/g creatinine women 20 men that persists at least 3 months. Generally, Cd-induced reductions eGFR are not reversible, nephropathy may progress end-stage disease. There evidence elimination current environmental exposure can reverse nephropathy, theoretical reason believe such reversal possible. Bioengineered in vitro models of the kidney offer unprecedented opportunities to better mimic vivo microenvironment. Kidney-on-a-chip technology reproduces 2D or 3D features which can replicate tissue architecture, composition, and dynamic mechanical forces experienced by cells vivo. Kidney are exposed stimuli such as substrate stiffness, shear stress, compression, stretch, regulate multiple cellular functions. Incorporating kidney-on-a-chip is critically important for recapitulating physiological pathological This review will explore approaches applying different cell types using how these systems used study physiology, model disease, screen drug toxicity. We further discuss sensor integration into monitoring responses other stimuli. advantages, limitations, challenges associated with incorporating a variety applications. Single-domain antibody (sdAb) is among the most promising vectors for developing molecular imaging tracers. Several sdAb tracers targeting human epidermal growth factor receptor 2 or programmed death ligand 1 have entered clinical practice. However, radiolabeled single-valent sdAbs generally high kidney retention, limiting their therapeutic applications. Therefore, engineering strategies such as PEGylation incorporation of renal cleavable linkers can be adapted to improve pharmacokinetics and reduce retention. In this Focus on Molecular Imaging review, we try summarize latest developments in sdAb-derived agents propose potential that used theranostic value sdAbs.

Ultrasmall gold nanoparticles (AuNPs) typically includes atomically precise nanoclusters (AuNCs) and AuNPs with a core size below 3 nm. Serving as bridge between small molecules traditional inorganic nanoparticles, the ultrasmall show unique advantages of both (e.g., rapid distribution, renal clearance, low non-specific organ accumulation) long blood circulation enhanced permeability retention effect). The emergence creates significant opportunities to address many challenges in health field including disease diagnosis, monitoring treatment. Since nano–bio interaction dictates overall biological applications AuNPs, this review elucidates recent advances interactions imaging AuNPs.
