Sirtuins in kidney diseases: potential mechanism and therapeutic targets

Abstract
Sirtuins,
which
are
NAD
+
-dependent
class
III
histone
deacetylases,
involved
in
various
biological
processes,
including
DNA
damage
repair,
immune
inflammation,
oxidative
stress,
mitochondrial
homeostasis,
autophagy,
and
apoptosis.
Sirtuins
essential
regulators
of
cellular
function
organismal
health.
Increasing
evidence
suggests
that
the
development
age-related
diseases,
kidney
is
associated
with
aberrant
expression
sirtuins,
regulation
sirtuins
activity
can
effectively
improve
delay
progression
disease.
In
this
review,
we
summarise
current
studies
highlighting
role
renal
diseases.
First,
discuss
sirtuin
family
members
their
main
mechanisms
action.
We
then
outline
possible
roles
cell
types
Finally,
compounds
activate
or
inhibit
consequently
ameliorate

Abstract
Microscopic

exosomes
are
leading
a
revolutionary
breakthrough
in
the
field
of
kidney
disease.
These
membrane‐enclosed
vesicles,
with
diameters
ranging
from
30
to
150
nm,
carry
cargo
such
as
proteins,
lipids,
and
nucleic
acids,
serving
bridge
information
between
cells
both
health
disease
states.
Recently,
significant
progress
has
been
made
understanding
critical
role
deciphering
diseases,
providing
novel
perspectives
on
mechanisms,
diagnosis,
monitoring,
prognosis,
treatment.
The
wide
range
sources
for
exosomes,
their
biocompatibility,
remarkable
stability,
well
convenient
storage
transport
capabilities,
make
them
rising
star
diagnostics.
immense
diagnostic
potential
harbored
within
these
tiny
structures
is
rapidly
emerging,
particularly
development
liquid
biopsy‐based
techniques.
This
article
provides
comprehensive
overview
biological
functions
disease,
highlighting
primary
markers.
Furthermore,
it
explores
applications
therapeutic
delivery,
paving
way
innovative
treatment
strategies.
Finally,
we
glimpse
into
future
research
prospects
this
field,
believing
that
continuous
advancement
exosome‐related
technologies,
diagnostics
will
become
more
precise
efficient,
bringing
hope
patients.

Despite
significant
advances
in
renal
physiology,
the
global
prevalence
of
chronic
kidney
disease
(CKD)
continues
to
increase.
The
emergence
multicellular
organisms
gave
rise
increasing
complexity
life
resulting
trade-offs
reflecting
ancestral
adaptations
changing
environments.
Three
evolutionary
traits
shape
CKD
over
lifespan:
1)
variation
nephron
number
at
birth,
2)
progressive
loss
with
aging,
and
3)
adaptive
growth
response
decreased
number.
Although
providing
plasticity
adaptation
environments,
cell
cycle
must
function
within
constraints
dictated
by
available
energy.
Prioritized
allocation
energy
through
placenta
can
restrict
fetal
nephrogenesis,
a
risk
factor
for
CKD.
Moreover,
aging
is
consequence
senescence,
pathway
accelerated
hypertrophy
that
maintains
metabolic
homeostasis
expense
increased
vulnerability
stressors.
Driven
reproductive
fitness,
natural
selection
operates
early
but
diminishes
thereafter,
leading
an
exponential

protect

phenotype
mitigate
physiological
cellular
senescence
buffer
stressors.

have

approaches
combat
adversity

evolutionary
process
provides
climate
change
confronts
rising
burden
accumulate

age.
