Environmental factors, including stress, that are experienced during early life (ELS) or adolescence potential risk factors for the development of behavioral and mental disorders later in life. The endocannabinoid system plays a major role regulation stress responses emotional behavior, thereby acting as mediator vulnerability resilience. Among critical which determine magnitude direction long-term consequences exposure is age, i.e., maturity brain circuits exposure. Thus, present study addressed hypotheses ELS adolescent differentially affect expression regulatory elements system, cannabinoid receptor 1 (CB1R) fatty acid amide hydrolase (FAAH) medial prefrontal cortex (mPFC) adult female rats. We also tested hypothesis proposed gene changes epigenetically modulated via altered DNA-methylation. specific aims were to investigate if (i) single stressors induce CB1R FAAH (ii) influences effect on expression, (iii) paralleled by DNA methylation. following experimental groups investigated: (1) non-stressed controls (CON), (2) (ELS), (3) (forced swimming; FS), (4) + FS found an up-regulation both single-stressor reduction back control levels group. An was only data indicate ELS, very immature stage development, exerts buffering programming induced stress. detected accompanied methylation patterns promoter region these genes, specifically, negative correlation mean with found. Our results induces “(re)programming” effect, characterized CpG-site within regions two genes influence response at adolescence.

Abstract Background Gut microbiota plays a pivotal role in the gut-brain axis and can influence neurodevelopment mental health outcomes. This review summarizes current evidence on associations between gut alterations various psychiatric illnesses. Main body The composition of microbiome evolves from birth through old age, disruptions during critical periods may increase disease risk. Factors like diet, medications, stress, infections disturb microenvironment lead to dysbiosis. Dysbiosis has been linked conditions depression, anxiety, autism, ADHD, schizophrenia. Proposed mechanisms involve microbial regulation neurotransmitters, inflammation, oxidative blood-brain barrier permeability, immune system. Therapeutic strategies probiotics, prebiotics, faecal transplantation modulate ecosystem. However, more research is needed elucidate causal microbiota-psychiatry relationship. Understanding interactions uncover new possibilities for preventing managing disorders. Conclusion A growing points close relationship health. While field still emerging, dysbiosis ecosystem associated with neuropsychiatric conditions. underlying likely microbiota-gut-brain signalling pathways. Objective Despite the overwhelming evidence for profound and longstanding effects of early‐life stress (ELS) on inflammation, brain structure, molecular aging, its impact human aging risk neurodegenerative disease is poorly understood. We examined ELS severity in interaction with age blood‐based markers neuroinflammation neurodegeneration, volumes, cognitive function middle‐aged women. Methods recruited 179 women (aged 30–60 years) without exposure before onset puberty. Using Simoa technology, we assessed including serum concentrations glial fibrillary acidic protein (GFAP) neurofilament light chain (NfL). further obtained T1‐weighted T2‐weighted magnetic resonance images to assess volumes performance sensitive early impairments associated development dementia, using Cambridge Neuropsychological Automated Test Battery. used generalized additive models examine nonlinear these outcomes. Results Analyses revealed significant NfL GFAP concentrations, total subcortical gray matter volume loss, increased third ventricular volume, impairment. Interpretation These findings suggest that profoundly exacerbates peripheral, neurostructural, aging. Our results are critical novel prevention strategies target developmental mitigate aging‐related neurological diseases. ANN NEUROL 2025

Suicide is the first cause of death among adolescents globally and has a severe impact on socioeconomic development. Several studies have found that suicide attempts suicidal ideation (SI) are more likely to occur in with depression. Previous stressful events early childhood, especially family stress, can influence occurrence SI. Chinese parents tend be strict less able express their emotions, owing unique national conditions, which may result parenting problems. Therefore, our study investigates risk factors for SI large sample A total 1604 adolescent patients depression were recruited this study. self-administered questionnaire collected clinical demographic data. was assessed by interview. Major depressive disorder (MDD) is an incapacitating condition characterized by loss of interest, anhedonia and low mood, which affects almost 4% people worldwide. With rising prevalence, it considered a public health issue that economic productivity heavily increases costs alone or as comorbidity for other pandemic non-communicable diseases (such obesity, cardiovascular disease, diabetes, inflammatory bowel diseases, etc.). What even more noteworthy the double number women suffering from MDD compared to men. In fact, this sex-related ratio has been contemplated since men have different sexual hormone oscillations, where meet significant changes depending on age range moment life (menstruation, premenstruation, pregnancy, postpartum, menopause…), seem be associated with susceptibility symptoms. For instance, decreased estrogen level promotes activation serotonin transporters. Nevertheless, hormones are not only triggers alter neurotransmission monoamines neuropeptides. Stress profoundly affects physical and mental health, particularly when experienced early in life. Early-life stress (ELS) encompasses adverse childhood experiences such as abuse, neglect, violence, or chronic poverty. These stressors can induce long-lasting changes brain structure function, impacting areas involved emotion regulation, cognition, response. Consequently, individuals exposed to high levels of ELS are at an increased risk for health disorders like depression, anxiety, post-traumatic disorders, well issues, including metabolic cardiovascular disease, cancer. This review explores the biological psychological consequences early-life adversity paradigms rodents, maternal separation deprivation limited bedding nesting. The study these experimental models have revealed that organism’s response is complex, involving genetic epigenetic mechanisms, associated with dysregulation physiological systems nervous, neuroendocrine, immune systems, a sex-dependent fashion. The amount of people affected by traffic-related air pollution and noise is continuously increasing, but limited research has been conducted on the association between these environmental exposures functional brain connectivity in children. This exploratory study aimed to analyze associations exposure during pregnancy childhood, amongst a wide-swath areas preadolescents from 9 12 years age. We used data 2,197 children Generation R Study. Land use regression models were applied estimate nitrogen oxides particulate matter levels at participant's homes for several time periods: pregnancy, birth 3 years, 6 age magnetic resonance imaging (MRI) assessment. Existing maps road traffic same periods. Resting-state MRI was obtained 9–12 Pair-wise correlation coefficients blood-oxygen-level-dependent signals 380 calculated. Linear regressions run corrected multiple testing. Preadolescents exposed higher NO2, NOx, PM2.5 absorbance, showed among regions (e.g. 0.16 0.19 coefficient related absorbance exposure, depending connection). Overall, most identified task positive negative networks, mainly inter-network (20 26). Slightly more than half connections intra-hemispheric (14 26), predominantly right hemisphere.
