Despite the advances in modern cancer therapy, malignant diseases are still a leading cause of morbidity and mortality worldwide. Conventional treatment methods frequently lead to side effects drug resistance patients, highlighting need for novel therapeutic approaches. Recent findings have identified existence non-canonical micropeptides, an additional layer proteome complexity, also called microproteome. These small peptides promising class agents with potential address limitations current treatments. The microproteome is encoded by regions genome historically annotated as non-coding, its has been revealed thanks recent proteomic bioinformatic technology, which dramatically improved understanding complexity. Micropeptides shown be biologically active several types, indicating their role. Furthermore, they characterized low toxicity high target specificity, demonstrating development better tolerated drugs. Non-coding RNAs (ncRNAs) have a significant role in gene regulation, especially cancer and inflammatory diseases. ncRNAs, such as microRNA, long non-coding RNAs, circular alter the transcriptional, post-transcriptional, epigenetic expression levels. These molecules act biomarkers possible therapeutic targets because aberrant ncRNA has been directly connected to tumor progression, metastasis, response therapy research. ncRNAs’ interactions with multiple cellular pathways, including MAPK, Wnt, PI3K/AKT/mTOR, impact processes like proliferation, apoptosis, immune responses. The potential of RNA-based therapeutics, anti-microRNA microRNA mimics, restore normal is being actively studied. Additionally, tissue-specific patterns ncRNAs offer unique opportunities for targeted therapy. Specificity, stability, responses are obstacles use ncRNAs; however, novel strategies, modified oligonucleotides delivery systems, developed. profiling may result more individualized successful treatments precision medicine advances, improving patient outcomes creating early diagnosis monitoring opportunities. current review aims investigate roles diseases, focusing on their mechanisms regulation implications non-invasive diagnostics therapies. A comprehensive literature was conducted using PubMed Google Scholar, research published between 2014 2025. Studies were selected based rigorous inclusion criteria, peer-reviewed status relevance Non-English, non-peer-reviewed, inconclusive studies excluded. Abstract Micropeptides are commonly identified as peptides encoded by non‐coding RNAs (ncRNAs). In the short open reading frame (sORF) of ncRNAs, there is a base sequence encoding functional micropeptides, which great significance in biological field. Recently, micropeptides regulate diverse processes, including mitochondrial metabolism, calcium transport, mRNA splicing, signal transduction, myocyte fusion, and cellular senescence, regulating homeostasis internal environment cancer's incidence progression. Especially, study cancer about potential regulatory mechanism will be conducive to further understanding process initiation development. More more research shows have been confirmed play an essential role emergence multiple kinds cancers, Breast cancer, Colon Colorectal Glioma, Glioblastoma, Liver cancer. Squalene epoxidase (SQLE) is a pivotal enzyme in sterol biosynthesis, catalyzing the conversion of squalene to 2,3-oxidosqualene. Beyond its core role cholesterol homeostasis, SQLE implicated cancer, hypercholesterolemia, and fungal infections, positioning it as valuable therapeutic target. We conducted comprehensive literature search across primary databases gather vitro, silico, vivo evidence on SQLE. This review explores enzyme's structural functional features, including substrate specificity catalytic mechanisms, examines inhibitor interactions. Computational methods predict - dynamics, guiding drug design, while investigations clarify SQLE's metabolic disorders tumorigenesis.
