Abstract Cortisol, also known as the stress hormone, is a crucial corticosteroid hormone that significantly increases secretion in human body when facing notable stress. Monitoring cortisol levels for personal management and diagnosis treatment of certain diseases. Electrochemical biosensors combine efficient sensitivity electrochemical technology with high specificity biological recognition processes, making them widely applicable analysis fluid components. This work outlines working mechanism biosensors, focusing particularly on sensing elements such antibodies, aptamers, molecularly imprinted polymers. It provides detailed explanations operational principles these different elements. summarizes evaluates latest advancements detecting fluids, discussing influence sensor design performance. Subsequently, through comparative various performances, further discusses challenges translating laboratory achievements into practical applications, including enhancing key metrics reusability, reproducibility, long‐term stability, continuous monitoring capability, response time. Exosomes, released from diverse cells as nanoscale lipid bilayer vesicles, mediate intercellular communication and participate in various physiological pathological processes. Thereinto, tumor-derived exosomes (T-EXOs) with molecular cargoes of parent tumor act attractive biomarkers for liquid biopsy. The amount T-EXOs their levels contained specific proteins nucleic acids are closely associated cancer burden classification. Nevertheless, the size relatively low abundance exosomes, well complex body matrix pose daunting challenges efficient isolation sensitive detection T-EXOs. Biosensing fast, convenient accurate method, has been widely employed over past decades. Among them, electrochemical sensors can sensitively detect by measuring change electrical signal caused oxidation or reduction at working electrode surface. This review aims to summarize recent advance biosensors quantification, protein RNA analysis exosomes. Further, future perspectives exosome-based biopsy have discussed.

Exosomes have been perceived as promising biomarkers for noninvasive cancer diagnosis and treatment monitoring. However, the sensitive accurate quantification phenotyping of exosomes remains challenging. Herein, a versatile electrochemiluminescence (ECL) aptasensor was proposed analysis tumorous exosomes. Specifically, ternary nanohybrid (Ru-HAuTiO2), by covalently linking ECL luminophore Ru(dcbpy)32+ with gold nanoparticles (AuNPs)-decorated hollow urchin-like TiO2 (HTiO2), ingeniously designed highly luminescent self-enhanced nanoemitter. Notably, porous HTiO2 played an "all-rounder" role, including carrier luminophores AuNPs, coreaction accelerator, specific exosome capturing scaffold through Ti–phosphate coordination interaction. On other hand, polydopamine modified covalent organic framework (PDA@COF) employed quencher to remarkably attenuate Ru-HAuTiO2 dual-quenching mechanism, further labeled aptamer (Apt) exosomal surface protein. Esophageal cancer is a common and lethal digestive system malignancy, both treatment efficacy patient survival rates face significant challenges. In recent years, exosomes have emerged as crucial mediators of intercellular communication, demonstrating tremendous clinical potential, particularly in the diagnosis, treatment, prognostic evaluation esophageal cancer. These not only serve biomarkers for early diagnosis prognosis but also modulate tumor growth, metastasis, drug resistance by delivering bioactive molecules. Importantly, can act carriers cancer-related therapeutic agents, optimizing gene therapy strategies to enhance while reducing toxicity side effects. Despite facing challenges applications such purification, enrichment, standardization analytical methods, maintain broad prospects application with potential significantly improve outcomes quality life.
