Abstract Long non-coding RNAs (lncRNAs) have emerged as crucial regulators in various cellular processes, including cancer progression and stress response. Recent studies demonstrated that copper accumulation induces a unique form of cell death known cuproptosis, with lncRNAs playing key role regulating cuproptosis-associated pathways. These may trigger cell-specific responses to stress, presenting new opportunities prognostic markers therapeutic targets. This paper delves into the cuproptosis-mediated cancer, underscoring their potential biomarkers targets for innovative strategies. Nanotherapy has emerged as a promising strategy for the targeted and efficient treatment of melanoma, most aggressive lethal form skin cancer, with minimized systemic toxicity. However, therapeutic efficacy cobalt oxide nanoparticles (Co3O4NPs) in melanoma remains unexplored. This study aimed to assess potential Co3O4NPs by evaluating their impact on cell viability, genomic DNA mitochondrial integrity, reactive oxygen species (ROS) generation apoptosis induction A-375 cells. Our findings demonstrated concentration-dependent reduction viability upon five Co3O4NP concentrations (0.2, 2, 20, 200, 2000 µg/ml), an IC50 value 303.80 µg/ml. Treatment this concentration significantly increased ROS generation, induced dramatic damage, disrupted membrane integrity. Flow cytometric analysis revealed necrosis following exposure at value. Lung adenocarcinoma (LUAD) is the most common subtype of lung cancer. Despite advances in treatment, prognosis remains poor due to late diagnosis. Cuproptosis (driven by copper ion accumulation) and disulfidptosis disulfide bond are novel forms programmed cell death, closely linked tumor initiation, progression, resistance. However, specific roles these mechanisms LUAD remain inadequately studied. This study integrated multi-omics data from TCGA GEO databases systematically evaluate differential expression prognostic significance disulfide-related genes (DCRGs), identify two DCRG molecular subtypes, construct a scoring model based on four key genes. We develop and evaluate copper-based metal-organic frameworks (Cu-MOFs) incorporating cromolyn as a linker to enhance structural stability, drug delivery efficiency, therapeutic potential, particularly for breast cancer treatment. Two Cu-MOF formulations were synthesized: Cu-MOFs-BDC-DOX (using terephthalic acid) Cu-MOFs-CROMO-DOX linker). Characterization was performed using SEM/TEM morphology, FTIR, XRD, TGA confirm integrity. Drug encapsulation efficiency release profiles assessed, followed by in vitro cytotoxicity, cell migration, colony formation assays MDA-MB-231 cells.
