Introduction: Ulcerative colitis (UC) is an inflammatory bowel disease influenced by genetic, immune, and environmental factors. This study investigates the link between ferroptosis, a cell death process related to oxidative stress iron metabolism, immune infiltration in UC. Materials Methods: We analyzed UC patient transcription data from Gene Expression Omnibus (GEO) identified ferroptosis-related genes using FerrDB. Using STRING Cytoscape, we protein-protein interactions identify hub Differentially Expressed Genes (UCDEGs) performed functional enrichment with GO KEGG pathways. Machine learning helped further key Ferroptosis-related (UCDE-FRGs), which were validated additional GEO datasets immunohistochemical staining. Results: A total of 11 UCDEGs ( CCL2, ICAM1, TLR2, CXCL9, MMP9, CXCL10, IL1B, CXCL8, PTPRC, FCGR3A , IL1A ) 3 UCDE-FRGs DUOX2, LCN2 IDO1 identified. indicates that these play role immunity ferroptosis. Analysis showed there large number Plasma cells, Monocytes, M0/M1 Macrophages Neutrophils Correlation analysis revealed associated immune-infiltrated cells IHC results expression levels all higher than healthy controls.
