Abstract The symbiotic bacteria that live in the human gut and metabolites they produce have long influenced local systemic physiological pathological processes of host. microbiota are increasingly being recognized for its impact on a range diseases, including cancer, it may play key role occurrence, progression, treatment, prognosis many types cancer. Understanding functional cancer is crucial development era personalized medicine. Here, we review recent advances research summarize important associations clear experimental evidence variety cancers, focus application possible challenges associated with antitumor therapy. In conclusion, our demonstrated multifaceted mechanisms affecting provides directions ideas future clinical research.

Accumulating evidence indicates that gut microbiota closely correlates with the tumorigenesis of digestive system cancers (DSCs). However, whether causality between and DSCs exists is unknown. Genome-wide association study (GWAS) summary statistics for bidirectional two-sample Mendelian randomization (MR) analysis were utilized to assess DSCs. Sensitivity analyses performed evaluate robustness our results. We found genus Eggerthella (OR = 0.464, 95%CI: 0.27 0.796, p 0.005) was negatively associated risk gastric cancer. The genetically predicted Lachnospiraceae FCS020 group 0.607, 0.439 0.84, 0.003) correlated a lower colorectal cancer, Turicibacter 0.271, 0.109 0.676, protective factor liver In reverse MR, regulated relative abundance specific strains microbiota. comprehensively screened using MR identified several microbial taxa Our discoveries are beneficial development novel markers microbiota-modifying therapeutics DSC patients.

Abstract In recent years, probiotics and their derivatives have been recognized as important therapeutic agents in the fight against cancer. Therefore, this study aimed to investigate anticancer effects of membrane vesicles (MVs) from Lentilactobacillus buchneri strain HBUM07105 probiotic isolated conventional unprocessed yogurt Arak province, Iran, gastric colon cancer cell lines. The MVs were prepared cell-free supernatant (CFS) L. characterized using field-emission scanning electron microscopy (FE-SEM) transmission (TEM) SPS-PAGE techniques. activity was evaluated MTT, flow cytometry, qRT-PCR techniques, a scratch assay. investigated anti-adenocarcinoma effect on human adenocarcinoma line (AGS) colorectal (HT-29) at 24, 48, 72-h time intervals. results demonstrated that all concentrations (12.5, 25, 50, 100, 200 µg/mL) reduced viability both types cells after 72 h treatment. analysis apoptosis revealed percentage AGS HT-29 early late stages significantly higher treatment compared untreated cells. After treating with MVs, arrested G0/G1 phase. These microvesicles demonstrate apoptotic by increasing expression pro-apoptotic genes ( BAX , CASP3 CASP9 ). According test, can decrease migration incubation control groups. also be considered potential option for inhibiting activities.

CRC remains a significant public health challenge due to its high prevalence and mortality rates. Emerging evidence highlights the critical role of gut microbiota in both pathogenesis efficacy treatment strategies, including chemotherapy immunotherapy. Dysbiosis, characterized by imbalances microbial communities, has been implicated progression therapeutic outcomes. This review examines intricate relationship between composition CRC, emphasizing potential for profiles serve as biomarkers early detection prognosis. Various interventions, such prebiotics, probiotics, postbiotics, fecal transplantation, dietary modifications, aim restore balance shift dysbiosis toward eubiosis, thereby improving Additionally, integration profiling into clinical practice could enhance diagnostic capabilities personalize advancing field oncology. The study intratumoral offers new prognostic tools that, combined with artificial intelligence algorithms, predict responses assess risk adverse effects. 3-hydroxybutyrate (3-HB), an essential endogenous metabolite, shows significant therapeutic potential in several disease contexts. However, its clinical application has been hampered by limitations, such as adverse effects on the gut microbiota. This study introduces a genetically engineered strain of Lactobacillus rhamnosus GG (LGGK) that integrates benefits 3-HB production with probiotic properties LGG. Using murine colon cancer cachexia (CAC) model, LGGK supplementation significantly improved survival, reduced tumor progression, and alleviated muscle wasting. restored microbial diversity, increased abundance beneficial bacteria, short-chain fatty acids while reducing harmful populations. In addition, demonstrated anti-inflammatory effects, effectively elevated pro-inflammatory cytokines serum skeletal muscle. These findings highlight dual-action approach utilizes metabolic gut-modulating innovation offers promising strategy for treatment CAC potentially other inflammatory disorders, highlights probiotics advanced applications.
