Senescence is irreversible cell cycle arrest that results from therapy-induced stress such as DNA damage. It was initially thought to be a tumor-suppressive mechanism, but now getting attention contribute tumor progression and therapy resistance through the senescence-associated secretory phenotype (SASP). Remodeling microenvironment (TME), SASP can establish conditions conducive progression. In addition, senescence being acknowledged increasingly crucial factor in inducing dormancy, state of reversible quiescence allows cancer cells evade therapeutic clearance survive protective niches. Eventually, both dormancy significantly maintenance stem (CSCs), enhancing their plasticity tumor-initiating potential. Moreover, promote aggressive disease cells, driving epithelial-to-mesenchymal transition (EMT) metastasis. On other hand, dormant act reservoir, serving seeds for metastatic spread which reactivate develop at secondary sites. Understanding mechanisms holds promise overcoming resistance, stemness Therapeutic strategies targeting include senolytics, senomorphics, dormancy-disrupting agents, immunotherapies. Future preclinical clinical research should prioritize integration senescence- dormancy-targeting agents with conventional treatments achieve durable control.

Despite their subordination in humans, to a great extent, mitochondria maintain independent status but tightly cooperate with the "host" on protecting joint life quality and minimizing health risks. Under oxidative stress conditions, healthy promptly increase mitophagy level remove damaged "fellows" rejuvenating mitochondrial population sending fragments of mtDNA as SOS signals all systems human body. As long metabolic pathways are under systemic control well-concerted together, adaptive mechanisms become triggered increasing protection, activating antioxidant defense repair machinery. Contextually, attributes patho-/physiology instrumental for predictive medical approach cost-effective treatments tailored individualized patient profiles primary (to protect vulnerable individuals again health-to-disease transition) secondary affected disease progression) care. Nutraceuticals naturally occurring bioactive compounds demonstrating health-promoting, illness-preventing, other health-related benefits. Keeping mind health-promoting properties nutraceuticals along therapeutic potential safety profile, there is permanently growing demand application mitochondria-relevant nutraceuticals. Application beneficial only if meeting needs at individual level. Therefore, risk assessment creation pivotal importance followed by adapted nutraceutical sets needs. Based scientific evidence available nutraceuticals, this article presents examples frequent which require protective measures targeted holistic following advanced concepts predictive, preventive, personalized medicine (PPPM/3PM)

Cellular senescence is a cell state triggered by programmed physiological processes or cellular stress responses. Stress-induced senescent cells often acquire pathogenic traits, including toxic secretome and resistance to apoptosis. When form faster than they are cleared the immune system, accumulate in tissues throughout body contribute age-related diseases, neurodegeneration. This review highlights evidence of brain their role Alzheimer's disease (AD), leading cause dementia older adults. We also discuss progress challenges senotherapies, pharmacological strategies clear mitigate effects, which hold promise as interventions for AD related dementias (ADRD).

Abstract Fisetin, a flavonol belonging to the flavonoid subclass, is ubiquitous dietary present in fruits and vegetables, including fruit peels, has proven potential for anticancer activity, especially lung cancer – leading cause of cancer-related deaths globally. The current paper provides most detailed elaborate list various roles fisetin experimentally induced cells, these include promotion apoptosis, inhibition cell proliferation, migration, invasion, as well regulation autophagy. Among molecular targets, some identified pathways, such PI3K/Akt, MAPK, NF-κB, that affects are crucial tumor formation, so it can be considered chemopreventive agent. Moreover, improves effectiveness conventional treatments chemo- radiosensitizer minimizes side effects. However, overall utility clinical use now somewhat restricted by its poor solubility short half-life. It predicted future development nanotechnologies drug delivery, nanoparticle encapsulation, might help solve difficulties. Further Preclinical investigations required uniformly determine safety, efficacy, standard dosage consumption therapy.
