Extracellular vesicles (EVs) possess the characteristics of their parent cells, based on which various studies have actively investigated treatments for diseases using mesenchymal stem cell-derived EVs due to regenerative activity. Furthermore, in recent years, there been significant efforts engineer improve native activities and integrate additional functions. Although both endogenous exogenous methods are used engineering EVs, may pose problem administering substances cells undergoing metabolic changes, can cause potential side effects. In addition, limitation losing beneficial factors inside membrane disruption during processes. Surface modification also impair efficiency presence proteins EV surface. Therefore, this study, a stable efficient method was achieved through ethanol-mediated hybridization functionalized lipid nanoparticles (LNPs) with fusogenic component. During hybridization, internal bioactive targeting moiety were maintained LNPs EVs. The Ab-Hybrid, successfully synthesized nicotinamide-encapsulated Col2A1 antibody-modified liposome Transforming growth factor-β1 (TGF-β1)-overexpressed administered osteoarthritis (OA)-induced rats destabilization medial meniscus surgery. Ultimately, Ab-Hybrid demonstrated excellent chondroprotective anti-inflammatory effects long-lasting properties OA lesions. We anticipate that approach manufacturing hybrid particles will serve as valuable versatile platform technology applicable diseases.
