ABSTRACT Molecular traces are increasingly being applied to assess the presence of species and communities. Studies on environmental DNA (eDNA) have, a large extent, become common practice in detection, but less studies have compared biodiversity estimations with more temporary RNA (eRNA). This study compares metabarcoding results from pond water obtained both molecule types by sequencing V4 region 18S rRNA marker. Water was collected two depths, 20 80 cm, filtered sequentially through filter porosities, 0.45 0.22 μm. Each cut half before fixation either 96% ETOH or RNAlater. The showed no differences between fixatives for molecule. Overall, estimates eDNA significantly overperformed eRNA, likely due higher concentrations terrestrial sources. Assessment of biodiversity using metabarcoding data, such as from bulk or environmental DNA sampling, is becoming increasingly relevant in ecology, sciences and monitoring. Thereby, the taxonomic identification species their sequences relies strongly on reference databases that link genetic to names. Microbial communities contain a broad phylogenetic diversity of organisms; however, the majority methods center on describing bacteria and archaea. In hot deserts daily/seasonal fluctuations pose great challenges to the resident organisms. However, these extreme ecosystems host unique microenvironments, such as rhizosheath-root system of desert speargrasses in which biological activities and interactions are facilitated by milder conditions reduced fluctuations. Currently there are sparse regulations regarding the discharge of antibiotics from wastewater treatment plants (WWTP) into river systems, making surface waters a latent reservoir for and antibiotic resistance genes (ARGs). To better understand factors that influence fate ARGs in environment to foster surveillance spreading such habitats, several indicator have been proposed, including integrase gene intI1 sulfonamide sul1 sul2.Here we used quantitative PCR long-read nanopore sequencing monitor abundance these present as class 1 integron cassettes system pristine source WWTP-impacted water. Therefore, limiting release ARG-harboring microorganisms may be more crucial restricting environmental antimicrobial than attenuating ng/L antibiotics.

Although many studies have shown that microbes can ectopically stimulate or suppress plant immune responses, the fundamental question of whether entire preexisting microbiota is indeed required for proper development response remains unanswered.
