Telomere length (TL) is considered a biomarker of ageing although this topic still debated. Also, sleep pattern changes are physiological part the normal process. In fact, it widely recognized that duration declines with age, leading to dysregulation circadian rhythms. The aim our study was analyse possible association TL in sample 135 subjects ages ranging from 20 111 years, recruited Palermo and neighbouring municipalities Sicily (Italy). Preliminary data suggest relative (RTL) decreases age both men women. However, at older ages, difference between women tends narrow. Nonagenarian centenarian do not show RTL values significantly different those observed adult old (40-89 years aged). Moreover, relationship sleep, we stratified into greater or lesser than 8-h periods. We found centenarians, who daily 8 hours more, have longer centenarians fewer hours. Although relatively small size provide preliminary evidence may affect centenarians. To best knowledge, first examine duration. Further studies required replicate extend these data.

Autoimmune diseases (ADs) are one of the groups chronic illnesses that impose a significant burden disease and health costs worldwide. Age is crucial risk factor for onset ADs. Theoretically, it inferred with organic immune system aging, loss tolerance specificity activity becomes more intense, probability autoimmunity increasing. However, there group individuals whose prevalence ADs very low or non-existent, despite biological aging. This paradox in raises questions. Centenarians, who over 100 years old, possibly most successful model aging humans. Most these exhibit favorable phenotype. To date, primary data evidence potential hypotheses explaining this phenomenon lacking globally, even though could provide valuable, original, relevant information regarding understanding protective factors, drivers, biomarkers related to autoimmunity. Herein we discuss some hypothesis may explain absence centenarians, including inflammaging, immunosenescence resilience, hyperstimulation, proteodynamics, genetics.

Several studies on the genetics of longevity have been reviewed in this paper. The results show that, despite efforts and new technologies, only two genes, APOE FOXO3A, involved protection cardiovascular diseases, shown to be associated with nearly all studies. This happens because genetic determinants are dynamic depend environmental history a given population. In fact, population-specific genes thought play greater role attainment than those shared between different populations. Hence, it is not surprising that GWAS replicated associations common variants few, if any, as these pool together An alternative way might study long-life families. type approach proving an ideal resource for uncovering protective alleles biological signatures healthy aging phenotypes exceptional longevity.

The proposed review aims to elucidate the intricate interplay between biological factors (sex differences) and socially constructed (gender in context of immune aging. While influence differences men women on various aspects responses has long been recognized, it is crucial acknowledge that gender, encompassing social cultural roles expectations associated with being male or female, also significantly shapes these processes. Gender can either accelerate aging promote longevity. By recognizing impact both factors, this work seeks offer a comprehensive understanding why may experience divergent trajectories varying outcomes terms Discrepancies perceived sexes, within families at work, contribute differing patterns antigen exposure. Additionally, variations micronutrient intake access preventive healthcare facilities exist. Health promotion knowledge often correlates educational attainment, which unequally represented males females many cultures across generations Western world. In countries without universal system, relies family prioritization strategies cope economic constraints, potentially limiting specific treatments affecting negatively. As result, behavioral gender disparities responses, susceptibility infections, autoimmune diseases, vaccine among older individuals. However, as demonstrated by COVID-19 pandemic, exhibit greater resilience infections than males. Given role system achieving longevity, not surprising live longer men, number female centenarians surpasses centenarians.

Polyunsaturated fatty acids (PUFA) are known to have a regulatory effect on oxidative and inflammatory processes. This study aimed identify the relationship between blood PUFA status circulatory markers of stress inflammation in cohort 172 subjects. The population was divided by sex into three age groups: adults (18-64 years old, n = 69), older (65-89 54), long-lived individuals (LLIs, 90-111 49). Whole content quantified using gas chromatography. Additionally, serum levels C-reactive protein (CRP), paraoxonase (PON), Trolox equivalent antioxidant capacity (TEAC), malondialdehyde (MDA) were measured. Our results showed that higher omega-3 (n-3) index adult females predictor lower MDA concentrations (p 0.038). Conversely, total n-3 n-6 positively related values among LLI men < 0.05), while inversely correlated with 0.05). Interestingly, increased TEAC 0.007), arachidonic acid (AA)/eicosapentaenoic (EPA) ratio 0.006). These findings suggest cellular is changes AA/EPA females, whereas may enhance men. Overall, our highlights complex, sex-specific interactions profiles across different groups.

Abstract The immunophenotype of oldest centenarians, i.e. semi- and supercentenarians, could provide important information about their ability to adapt factors associated with immune changes, including ageing per se chronic Cytomegalovirus infection. We investigated, by flow cytometry, variations in percentages absolute numbers cell subsets, focusing on T cells, pro-inflammatory parameters a cohort 28 women 26 men (age range 19–110 years). observed variability hallmarks immunosenescence related age serological status. eight centenarians showed the lowest naïve due age, highest T-effector memory cells re-expressing CD45RA (TEMRA), according cytomegalovirus status, high levels serum parameters, although means were lower than that remaining 90+ donors. Some them CD8 TEMRA percentages, exhaustion/pro-inflammatory markers comparable younger ones. Our study supports suggestion ageing, especially exhibits great is not only attributable single contributor but should also be full result combination several factors. Everyone ages differently because he/she unique genetics experience life this applies even more system; everybody has had different immunological history. Furthermore, our findings inflammatory markers, CMV seropositivity discussed light most recent literature, suggest these changes might unfavourable for particular

Human ageing can be characterized by a profile of circulating microRNAs (miRNAs), which are potentially predictors biological age. They used as biomarker risk for age-related inflammatory outcomes, and senescent endothelial cells (ECs) have emerged possible source miRNAs. In this paper, panel four miRNAs including miR-146a-5p, miR-126-3p, miR-21-5p, miR-181a-5p, involved in several pathways related to inflammation, ECs senescence that seem characteristic the healthy phenotype. The levels these were determined 78 subjects aged between 22 111 years. Contextually, extracellular miR-181a-5p measured human vitro model, undergoing senescence. We found miRNAs, using ex vivo models, progressively increase with age, apart from ultra-centenarians showed comparable those young individuals. Our results contribute development knowledge regarding identification biomarkers successful unsuccessful ageing. Indeed, they might diagnostic/prognostic relevance diseases.

The immune system of semi- and supercentenarians (i.e., the oldest centenarians) is believed to have peculiar characteristics that enable them reach extreme longevity in a relatively healthy state. Therefore, previous papers, we investigated, through flow cytometry, variations percentages main subsets Tαβ Tγδ cells Sicilian cohort 28 women 26 men (age range 19-110 years), including 11 long-living individuals (>90 years old) 8 centenarians. These investigations suggested some observed immunophenotypic changes may contribute In present study, further characterize immunophenotype centenarians, examined Natural Killer (NK) identified as CD3-CD56 + CD16+ previously described cohort. We found highly significant increase NK cell with age. When stratified by gender, this age was maintained both sexes, higher significance males. Our findings on cells, together obtained results, discussed context literature, suggest these are not unfavourable for ones, supporting hypothesis aging should be considered differential adaptation rather than general alteration. adapted mechanisms allow centenarians successfully adapt history insults achieve remarkable longevity.
