Intracellular density impacts the physical nature of cytoplasm and can globally affect cellular processes, yet regulation remains poorly understood. Here, using a new quantitative phase imaging method, we determined that dry-mass in fission yeast is maintained narrow distribution exhibits homeostatic behavior. However, varied during cell cycle, decreasing G2, increasing mitosis cytokinesis, dropping rapidly at birth. These variations were explained by constant rate biomass synthesis, coupled to slowdown volume growth division rapid expansion post-cytokinesis. Arrest specific cell-cycle stages exacerbated changes. Spatially heterogeneous patterns suggested links between regulation, tip growth, intracellular osmotic pressure. Our results demonstrate systematic cycle are predominantly due modulation expansion, reveal functional consequences gradients arrests.

Fission yeasts are an ancient group of fungal species that diverged from each other tens to hundreds million years ago. Among them is the preeminent model organism Schizosaccharomyces pombe, which has significantly contributed our understandings molecular mechanisms underlying fundamental cellular processes.
