so

Abstract
Background
Traditional
Chinese
medicine
(TCM)
has
found
widespread
application
in
neoplasm
treatment,
yielding
promising
therapeutic
candidates.
Previous
studies
have
revealed
the
anti-cancer
properties
of
Brevilin
A,
a
naturally
occurring
sesquiterpene
lactone
derived
from
Centipeda
minima
(L.)
A.Br.
(
C.
),
TCM
herb,
specifically
against
lung
cancer.
However,
underlying
mechanisms
its
effects
remain
elusive.
This
study
employs
network
pharmacology
and
experimental
analyses
to
unravel
molecular
A
Methods
The
Batman-TCM,
Swiss
Target
Prediction,
Pharmmapper,
SuperPred,
BindingDB
databases
were
screened
identify
targets.
Lung
cancer-related
targets
sourced
GEO,
Genecards,
OMIM,
TTD,
Drugbank
databases.
Utilizing
Cytoscape
software,
protein-protein
interaction
(PPI)
was
established.
Gene
Ontology
(GO),
Kyoto
Encyclopedia
Genes
Genomes
(KEGG),
set
enrichment
analysis
(GSEA),
gene-pathway
correlation
conducted
using
R
software.
To
validate
results,
docking,
dynamics
simulations,
vitro
experiments
performed.
Results
We
identified
599
A-associated
3864
targets,
with
155
overlapping
genes
considered
as
candidate
for
PPI
highlighted
STAT3,
TNF,
HIF1A,
PTEN,
ESR1,
MTOR
potential
GO
KEGG
2893
enriched
terms
157
pathways,
including
PI3K-Akt
signaling
pathway,
FoxO
HIF-1
pathway.
GSEA
demonstrated
close
association
between
hub
Gene-pathway
indicated
significant
associations
cellular
response
hypoxia
Molecular
docking
simulations
confirmed
A's
PTEN
respectively.
In
A-induced
dose-
time-dependent
cell
death
A549
cells.
Notably,
treatment
significantly
reduced
HIF-1α
mRNA
expression
while
increasing
levels.
Conclusions
elucidates
