Doxorubicin (Dox) is an effective and commonly used anticancer drug; however, it leads to several side effects including cardiotoxicity which contributes poor quality of life for cancer patients. Creatine (Cr) a promising intervention alleviate Dox-induced cardiotoxicity. This study aimed examine the Cr beforeDox on cardiac mitochondrial creatine kinase (MtCK). Male rats were randomly assigned one two 4-week feeding interventions (standard diet or loading diet) control (Con, n = 20). Rats in standard (Cr1, 20) fed 2% 4-weeks. (Cr2, 4% 1-week followed by 3-weeks. After 4-weeks, received either bolus injection 15 mg/kg Dox placebo saline (Sal). Five days post-injections left ventricle (LV) was excised analyzed MtCK expression using Western blot ELISA. A significant drug effect observed LV mass (p < 0.05), post hoc testing revealed Con + Cr2 significantly lower than Sal 0.05). 0.03) through blot. interaction 0.02) Post that had higher Cr1 Sal. Data suggest reduction may contribute cardiotoxicity, supplementation play potential role mitigating preserving CK.

The aim of this study was to investigate AMD1 cardiotoxicity function for Maduramicin (Mad). SD rats were divided into control (Control) group and Mad treatment (3.5 mg/kg) After with seven days, the levels LDH CK-MB in serum detected, H&E staining TUNEL performed. In vitro, 1.0 μm used subsequently experiment, observing cell apoptosis from Flow cytometry. Caspase-3 detected Western blotting. cytometry blotting also performed after use siRNA-AMD1-1. Then, analysis potential bioinformatics techniques including GO, KEGG, PPI, immune infiltration molecular docking. has myocardial toxic effects vivo which raised. When knocked down, alleviated. Apoptosis, proliferation inflammation major pathophysiological changes process AMD1-knockdown. This involved IL1A, IL1B, PTGS2, VEGFA, VEGFC HBEFG, as hub genes related Maduramicin. their microenvironment changes: Effector memory CD4 T Natural killer more infiltrated, Mast less infiltrated. exerted cardiotoxic by upregulating gene, may be associated apoptosis, inflammatory response. had function, impact both cells they live.
