The primary objective of this systematic review study was to investigate the effectiveness, durability, and adverse events PLLA treatment for aesthetic indications. search strategy performed in MEDLINE (Ovid). electronic literature five databases performed, from inception until 12th February 2024. This identify randomized clinical trials that assessed adult individuals exhibiting facial aging and/or lipoatrophy. Risk bias using Cochrane Risk-of-Bias Tool Randomized Trials (RoB 2). Eleven RCTs out 1467 identified citations were included. Four studies showed increased dermal thickness, significant improvement lipoatrophy severity scores, after with its effects sustained at least 25 months. Two demonstrated superiority over injectable human collagen. Also, three positive results favoring when compared PH gel severity, transepidermal water loss, skin quality, elasticity, patient satisfaction. All mild-to-moderate intensity, main ones worth noting bruising, hematoma, tenderness, nodules, edema. Five eleven considered having high risk bias. evidence on effectiveness safety rejuvenation is low quality; thus, reported safety, long-lasting purpose should be further investigated.

ABSTRACT Background Calcium hydroxyapatite (CaHA) fillers are widely used for soft‐tissue augmentation due to their volumizing properties and biostimulatory effects. However, clinical evidence supporting the dual role of CaHA as both a volumizer skin booster remains limited. Objectives To evaluate efficacy filler (VoLassom) in midface volume restoration assess its effects on barrier function, hydration, elasticity over 24 weeks. Methods Fifteen participants aged 19–70 years with moderate deficiency (Midface Volume Deficiency Scale [MFVDS] grade ≥ 3) received injections. Clinical assessments included MFVDS scoring, Global Aesthetic Improvement (GAIS), patient satisfaction ratings at baseline weeks 2, 4, 24. Instrumental evaluations transepidermal water loss (TEWL), corneometer‐based cutometer‐measured (R2, R5, R7) were conducted all timepoints. Results scores significantly decreased from 24, demonstrating sustained restoration. GAIS remained high, rated improved (5.0) 2 though slightly reduced week (4.53 ± 0.52). Patient consistently high. TEWL 4 ( p < 0.01), indicating function. Corneometer measurements showed significant hydration improvements timepoints 0.05 0.001, respectively). Skin elasticity, measured by R2, R7, increased suggesting enhanced firmness. Conclusions demonstrated effective while improving elasticity. These findings suggest that functions not only but also booster, offering comprehensive approach facial rejuvenation. Nevertheless, further studies larger cohorts warranted validate these results.

Poly-L-Lactic Acid (PLLA) is a synthetic polymer which possesses biocompatible and biodegradable properties, widely used in the clinical filler material. This study focuses on potential role of PLLA collagen production dermal fibroblasts its mechanism. The fibroblast Hs60 was treated with different concentration PLLA. RT-qPCR conducted for determination mRNA levels type I (COL1) alpha 1 (COL1A1), COL1 2 (COL1A2), elastin, matrix metalloproteinase (MMP-1), tissue inhibitor (TIMP-1), TIMP-2. Procollagen Type C-peptide (PIP) enzyme immunoassay (EIA) Kit assay carried out to analyze procollagen production. Western Blot employed examine effect transforming frown factor (TGF-β) receptor-specific (SB431542) protein COL1A1 TGF-β/Smad signaling pathway related proteins. With increase concentration, gradually increased, both COL1A2 increased (p < 0.001). Elevated concentrations TIMP-1, TIMP-2 attenuated MMP-1 expression. TGF-β dose-dependently manner. p-Smad2 p-Smad3 were also by PLLA, but influences reversed SB431542 Similarly, COL1A1, COL1A2, caused significantly inhibited SB431542, whereas typically elevated 0.001).Poly-L-Lactic promotes activating pathway. The findings may lay foundation material applications
