Objective Lung cancer, the most prevalent malignancy, is typically diagnosed at an advanced stage. Smoking a pivotal risk factor for NSCLC, yet impact of various smoking statuses on NSCLC remains unclear. Thus, this study aims to explore whether different can causally influence through effects predictive targets, offering novel perspective treatment. Methods Employing dual-sample MR, MVMR, and TSMR approaches, we assessed causal relationships between 13 distinct using predicted potential therapeutic targets as mediators further elucidate interplay among them. Results Among statuses, current tobacco smoking, exposure smoke outside home, past never smoked demonstrated with NSCLC. MVMR analysis reveals that Current independent Utilizing NCAPD2, IL11RA, MLC1 mediators, IL11RA (22.2%) was found potentially mediate relationship Conclusion This study, integrating bioinformatics MR analysis, identified three investigate providing new insights treatment prevention

This study employed Mendelian Randomization (MR) to investigate the causal relationships among immune cells, COPD, and potential metabolic mediators. Utilizing summary data from genome-wide association studies, we analyzed 731 cell phenotypes, 1,400 plasma metabolites, COPD. Bidirectional MR analysis was conducted explore links between cells complemented by two-step mediation multivariable identify mediating metabolites. Causal were identified 41 phenotypes with 6 exhibiting reverse causality. Additionally, 21 metabolites causally related Through analyses, 8 found have COPD mediated (including one unidentified), 1-methylnicotinamide levels showing highest proportion at 26.4%. We These findings contribute screening of individuals high risk for offer insights into early prevention precocious diagnosis Pre-COPD.

Recently, numerous studies have explored the potential impact of gut microbiota on Chronic Obstructive Pulmonary Disease (COPD). However, causal relationship between skin and COPD, as well differences similarities relationships has not been thoroughly studied. We conducted a comprehensive two-sample Mendelian randomization (MR) analysis to investigate COPD. The inverse variance weighted (IVW) method was used primary approach. MR-Egger, median, MR-PRESSO methods were supplementary approaches. Various sensitivity stability analyses validate results. Genetic variations obtained from FR02 cohort study. derived KORA FF4 PopGen cohorts, with total 1,656 samples. GWAS data for COPD FinnGen consortium, including 18,266 cases 311,286 controls European cohorts. results IVW MR showed that 10 microbiotas 4 negatively associated [p < 0.05, odds ratio (OR) 1]; 3 6 positively (p OR > 1). None them heterogeneous or horizontally pleiotropic 0.05) reverse causality. This study revealed offering fresh perspectives prevention, diagnosis, management

Studies have shown a close correlation among immune cells, plasma metabolites, and atrial fibrillation (AF). However, it is not clear if this association related, which we used Mendelian randomization (MR) to investigate. We analyzed the between AF by using summarized data from genome-wide studies. Among them, explored associations cells bidirectional MR analysis. Combined with mediation analysis multivariable MR, further identified potential mediating plasmic metabolites. Results shows that causal relationships 8 cell phenotypes were all exhibiting reverse causality. Furthermore, 22 metabolites relationship AF. In addition, 2 including CD25 on IgD + CD38dim CX3CR1 CD14 CD16-monocyte, found 4 4-acetamidobutanoate levels, Octadecanedioylcarnitine (C18-DC) Linolenate [alpha or gamma; (18:3n3 6)] N-acetyl-aspartyl-glutamate might be mediators. Ultimately, only CD38dim, did appear function as mediators ( P -value = .030 < .05). conclusion, are causally associated levels mediate pathway linking This finding provides new perspective for early prevention diagnosis of preatrial

Objective Dietary factors and nutritional status may be among the risk for Chronic Obstructive Pulmonary Disease (COPD). There exists a certain correlation between trace elements COPD. Through Mendelian Randomization (MR) analysis, we investigated causal relationships elements, inflammatory proteins, Methods We employed MR, multivariable MR (MVMR), two-step (TSMR) approaches to assess links 15 COPD, with 91 proteins serving as mediators further elucidate tripartite relationships. Results Trace such Folate (OR = 1.293, 95%CI 1.027–1.628; p 0.029), Vitamin D 1.331, 1.071–1.654; 0.010), B12 1.424, 1.108–1.828; 0.006), Iron 0.741, 0.580–0.946; 0.016) demonstrated No relationship was observed in reverse MR. After adjusting BMI, 1.633, 1.098–2.429; 0.015), 0.507, 0.31–0.778; 0.001), 1.511, 1.029–2.217; 0.034) were identified independent whereas 1.118, 0.751–1.666; 0.581) not. Mediation analysis indicated that CDCP1 (5.76%) play mediating role Conclusion Folate, D, B12, have BMI adjustment, emerge factors. Furthermore, protein partially mediate offering scientific basis dietary recommendations could benefit COPD patients. The supplementation of advantageous individuals suffering from
