Anakinra and canakinumab are two FDA-approved IL-1 blockers indicated for the treatment of multiple autoinflammatory diseases, yet their safety has not been comprehensively analyzed. We aimed to assess signals associated with anakinra by conducting a pharmacovigilance analysis using FDA Adverse Event Reporting System (FAERS) database. reaction data spanning from first quarter 2004 fourth 2023 was downloaded FAERS A disproportionality utilizing various methods, including reporting odds ratio (ROR), proportional (PRR), Bayesian confidence propagation neural network (BCPNN), Empirical Bayes Geometric Mean (EBGM), conducted. were identified as primary suspect drugs adverse events (AEs) in 7,544 8,044 reports, respectively. The most commonly reported SOCs both general disorders administration site conditions. Subgroup analyses that SOC among health professionals non-health remained consistent across medications. At preferred term (PT) level, drug labeling, common AEs included injection reactions (ISRs) infections. Further revealed higher frequency ISRs anakinra, pain erythema. In contrast, more gastrointestinal (abdominal pain, mouth ulceration, inflammatory bowel disease) respiratory (cough, oropharyngeal rhinorrhea); these conditions predominantly occurred minors. Notably, no significant related tuberculosis infection or reactivation observed, hepatic injury malignancy low. This study confirms favorable profiles canakinumab, offering critical insights into rational usage regulations.

ABSTRACT The expression levels of macrophage‐associated cytokines are significantly greater in COVID‐19 patients than healthy individuals. Exploring strategies to modulate pathological cytokine storms can effectively prevent the development severe coronavirus infection‐induced pneumonia. Treatment with interleukin‐37 (IL‐37), an anti‐inflammatory factor, has unique and antiviral effects on infections caused by various pathogens. In this study, we investigated effect IL‐37 treatment SARS‐CoV‐2 Omicron‐infection induced inflammatory response its molecular mechanism.
