Objective A feared complication of aminoglycoside treatment is ototoxicity, which theorized to be attributed the production aminoglycoside‐induced reactive oxygen species. Previous studies using animal models have suggested that numerous therapies targeting reducing oxidative stress may prevent ototoxicity from aminoglycosides. However, few clinical been conducted on these antioxidants. This systematic review and meta‐analysis examines effectiveness antioxidant in ototoxicity. Data Sources PubMed, Embase, Web Science, ClinicalTrials.gov . Review Method literature search was August 2024. sought randomized controlled trials humans examining otologic outcomes following administration medications intended reduce stress. Results yielded 2037 results, seven met inclusion criteria. N‐acetylcysteine (NAC) investigated four studies, aspirin two vitamin E one study. Ischemic stroke poses a major threat to human health. Therefore, the molecular mechanisms of cerebral ischemia/reperfusion injury (CIRI) need be further clarified, and associated treatment approaches require exploration. The NOD‑like receptor thermal protein domain 3 (NLRP3) inflammasome serves an important role in causing CIRI, its activation exacerbates underlying injury. Activation NLRP3 triggers maturation production inflammatory molecules IL‑1β IL‑18, as well gasdermin‑D‑mediated pyroptosis CIRI damage. Thus, may viable target for CIRI. In present review, intense response induced by are discussed, therapeutic strategies that NLRP3‑mediated summarized. At present, certain drugs have already been studied, highlighting future perspectives.

Oxidative stress, characterized by an imbalance between excessive reactive oxygen species (ROS) production and impaired antioxidant defenses, is closely linked to the pathogenesis of various otorhinolaryngological disorders. Mitochondria, as primary site cellular energy production, play a crucial role in modulating oxidative stress. Mitochondrial dysfunction exacerbates ROS generation, leading damage inflammatory responses. In diseases, stress strongly associated with conditions such hearing loss, allergic rhinitis, chronic sinusitis, where tissue inflammation are key pathological features. Recent studies have highlighted potential therapies mitigate restore homeostasis, offering promising avenues for alleviating symptoms these diseases. However, despite encouraging results from early-stage research, clinical efficacy interventions remains be fully established. This review provides overview diseases evaluates therapeutic strategies.

Sensorineural hearing loss (SNHL) is a category of that often leads to difficulty in understanding speech and other sounds. Auditory system dysfunction, including deafness auditory trauma, results cognitive deficits via neuroplasticity. Cognitive impairment (CI) refers an abnormality the brain’s higher intellectual processes related learning, memory, thinking judgment can lead severe learning memory deficits. Studies have established strong correlation between SNHL CI, but it remains unclear how contributes CI. The purpose this article describe three hypotheses regarding relationship, mainstream load hypothesis, co-morbidity sensory deprivation as well latest research progress each hypothesis.

Reactive oxygen species (ROS) were formerly known as mere byproducts of metabolism with damaging effects on cellular structures. The discovery and description NADPH oxidases (Nox) a whole enzyme family that only produce this harmful group molecules was surprising. After intensive research, seven Nox isoforms discovered, described extensively studied. Among them, the oxidase 3 is perhaps most underrated isoform, since it firstly discovered in inner ear. This stigma Nox3 "being expressed ear" also used by me several times. Therefore, question arose whether sentence still valid or even usable. To end, review solely focuses summarizes its discovery, structural components, activating regulating factors, expression cells, tissues organs, well beneficial detrimental Nox3-mediated ROS production body functions. Furthermore, involvement Nox3-derived diseases progression and, accordingly, potential target for disease treatment, will be discussed.

Gravitational changes have been shown to cause significant abnormalities in various body systems, including the cardiovascular, immune, vestibular, and musculoskeletal systems. While numerous studies examined response of vestibular system gravitational stimulation, research on functional peripheral inner ear remains limited. The comprises two closely related structures: vestibule cochlea. These components share similar structures neural functions, highlighting importance investigating auditory nerve cells alterations. To address this gap, we studied structural following exposure hypergravity stimuli. Our findings demonstrate brainstem responses (ABRs) ABR recordings were used analyze click thresholds, as well amplitude latency tone bursts. thresholds at all frequencies increased group exposed long term. Additionally, burst results revealed significantly reduced amplitudes high delayed latencies models. Notably, greater hair cell loss was observed middle basal turns cochlea, indicating that mid high-frequency regions are more vulnerable stimulation. Furthermore, damage cochlear surface evident subjects 4G stimulation for 4 weeks. suggest its activity can be functionally structurally affected by prolonged hypergravity.

Abstract Noise‐induced hearing loss (NIHL) results from prolonged exposure to intense noise, causing damage sensory outer hair cells (OHCs) and spiral ganglion neurons (SGNs). The blood labyrinth barrier (BLB) hinders systemic drug delivery the inner ear. This study applied a retro‐auricular round window membrane (RWM) method bypass BLB, enabling transport of macromolecular proteins into Pigment epithelium‐derived factor (PEDF), which has anti‐inflammatory neuroprotective properties, is conjugated prestin‐targeting peptide 2 (PrTP2) using N‐succinimidyl‐3‐maleimidopropionate (SMP) form PrTP2‐SMP/PEDF. compound specifically targeted Prestin accumulated around OHCs for sustained release, effectively reducing OHC SGN loss. Functional structural tests, including auditory brainstem response (ABR), confocal microscopy, scanning electron microscopy (SEM), revealed significant restoration cellular protection. Additionally, enzyme‐linked immunosorbent assay (ELISA), Annexin V propidium iodide (PI) staining immunoblotting show that noise may induce pyroptosis in cochlea by activating NOD‐like receptor protein 3 (NLRP3)‐apoptosis‐associated speck‐like containing CARD (ASC) – cysteinyl aspartate specific proteinase (Caspase‐1) pathway PrTP2‐SMP/PEDF alleviates inflammatory inhibiting pyroptosis. Toxicity analysis indicates no adverse effects, suggesting promising therapeutic prospective NIHL.

This study aimed to investigate the relationship between neutrophil-to-HDL cholesterol ratio (NHR) and risk of hearing loss, as well evaluate potential NHR a biomarker for loss. The U.S. National Health Nutrition Examination Survey (NHANES) data covering 2005-2012 2015-2020 were analyzed. A weighted multivariate logistic regression model assessed correlation speech-frequency loss (SFHL) high-frequency (HFHL). Restricted cubic spline (RCS) analysis was utilized nonlinear correlation. Additionally, subgroup performed identify differences among subgroups. receiver operating characteristic (ROC) conducted efficacy in predicting total 10,436 participants involved. After comprehensive adjustments confounding factors, linearly correlated with SFHL HFHL. Subgroup revealed that race poverty index (PIR) significantly modified association ROC demonstrated predictive capability is positively suggests may serve assessing demonstrating significant clinical application value. However, this cross-sectional limits ability establish causality. Future longitudinal studies are needed confirm these findings explore mechanisms.
