Natural antioxidants represented by quercetin have been documented to be effective against atherosclerosis. However, the related mechanisms remain largely unclear. In this study, we identified a novel anti-atherosclerotic mechanism of inhibiting macrophage pyroptosis activating NRF2 through binding Arg483 site KEAP1 competitively. ApoE-/- mice fed with high fat diet, administration attenuated atherosclerosis progression reducing oxidative stress level and suppressing pyroptosis. At cellular level, suppressed THP-1 induced ox-LDL, demonstrated NLRP3 inflammasome activation ROS while these effects were reversed specific inhibitor (ML385). Mechanistically, promoted dissociate from KEAP1, enhanced nuclear translocation as well transcription downstream antioxidant protein. Molecular docking results suggested that could bind at Arg415 Arg483. order verify sites, mutated Ser (R415S R483S) was transfected into macrophages, anti-pyroptotic effect abrogated mutation, but not mutation. Furthermore, after adeno associated viral vector (AAV) AAV-KEAP1-R483S, almost abolished in mice. These findings proved quercetins targeting KEAP1/NRF2 interaction, provided reliable data on underlying natural protect

Lifestyle is an important factor in health, and researchers are trying to see the adverse effect on human health of a change lifestyle. Many people follow unhealthy lifestyles, which affect physical mental health. Millions suffer from various problems like cardiovascular disease, overweight, malnutrition, hypertension, stress, violence, etc. There many medicines that can cure these diseases, but they have side effects. When man consumes green leaves vegetables his diet, chances reducing risk any disease increase. Every plant has primary secondary metabolites. Flavonoid metabolite found plants plays role survival by performing physiological activities being able withstand effects environment. Quercetin one such bioflavonoid plants, onions, citrus fruits, parsley, It comes under category flavone, with five hydroxy groups chromen-4-one skeletal structure. applications, include anti-cancer, anti-bacterial, anti-viral, anti-inflammatory, anti-diabetic, However, use quercetin limited due its poor bioavailability, aqueous solubility, lower stability gastrointestinal tract, first-pass effect. To enhance application quercetin, derivatives metal complex required water soluble, helps them reach target site without provides stability. This review divided into three main sections: The first section explains about flavonoids their types; second methods synthesis, derivatives, complex; last represents complete applications drawbacks how overcome or deal drawbacks.

Quercetin, as a member of flavonoids, has emerged potential therapeutic agent in cardiovascular diseases (CVDs) recent decades. In this comprehensive literature review, our goal was critical appraisal the pathophysiological mechanisms quercetin relation to classical risk factors (e.g., hyperlipidemia), atherosclerosis, etc. We also assessed experimental and clinical data about its application CVDs. Experimental studies including both vitro methods vivo animal models mainly outline following effects quercetin: (1) antihypertensive, (2) hypolipidemic, (3) hypoglycemic, (4) anti-atherosclerotic, (5) cardioprotective (suppressed cardiotoxicity). From point view, there are human meta-analyses implicating beneficial on glycemic lipid parameters. contrast, other failed demonstrate consistent favorable cardiometabolic such MS, obesity, hypertension, underlying need for further investigation. Analyzing reason inconsistency, we identified significant drawbacks trials’ design, while absence pharmacokinetic/pharmacodynamic tests prior attenuated power results. Therefore, additional well-designed preclinical required examine efficacy

Abstract Resveratrol, curcumin, and quercetin are the secondary metabolites from medicinal food homology plants, that have been proven their potency in cancer treatment. However, antitumor effect of a single component is weak. So, herein, we designed an compound named RCQ composed resveratrol, quercetin. This study examined on tumorigenesis development 4T1 breast cancer-bearing mice following administering by intragastric administration. increased recruitment T cells reduced accumulation neutrophils macrophages tumor microenvironment. Meanwhile, suppressed tumor-infiltrating lymphocytes into immunosuppressive cell subpopulations, including CD4 + to helper Type 2 type (Th2), tumor-associated (TANs) N2 TANs, (TAMs) M2 TAMs. reversed predominance infiltrating microenvironment tipped immune balance toward activation state. In vitro showed significantly reactive oxygen species (ROS), reduce mitochondrial membrane potentials cells, modulate pro-apoptotic Bcl-2 family members. conclusion, can promote ROS apoptosis mechanism alleviate immunosuppression enhance anti-tumor effect.

Alzheimer's disease (AD) is a major global health problem today and the most common form of dementia. AD characterized by formation β-amyloid (Aβ) plaques neurofibrillary clusters, leading to decreased brain acetylcholine levels in brain. Another mechanism underlying pathogenesis abnormal phosphorylation tau protein that accumulates at level aggregates, areas affected this pathological process are usually cholinergic neurons cortical, subcortical, hippocampal areas. These effects result cognitive function, atrophy, neuronal death. Malnutrition weight loss frequent manifestations AD, these also associated with greater decline. Several studies have confirmed balanced low-calorie diet proper nutritional intake may be considered important factors counteracting or slowing progression whereas high-fat hypercholesterolemic predisposes an increased risk developing AD. Especially, fruits, vegetables, antioxidants, vitamins, polyunsaturated fatty acids, micronutrients supplementation exert positive on aging-related changes due their antioxidant, anti-inflammatory, radical scavenging properties. The purpose review summarize some possible contribute prevention understand role nutrition plays Aβ typical neurodegenerative disease, identify potential therapeutic strategies involve natural compounds, delaying disease.

Quercetin is a representative flavonoid that widely present in fruits, herbs, and vegetables. It also an important active core component traditional Chinese medicines. As flavonoid, quercetin has various properties exerts antioxidant, anti-inflammatory, cardioprotective effects. The public interest increasing, been used to prevent or treat numerous of diseases, such as polycystic ovary syndrome (PCOS), cancer, autoimmune diseases chronic cardiovascular clinical experiments animal studies due its powerful antioxidant minimal side marked pharmacological effects on gynecological disorders; however, there have no reviews about the potential health benefits context disorders, including PCOS, premature failure (POF), endometriosis (EM), ovarian cancer (OC), cervical (CC) endometrial carcinoma (EC). Thus, this review aimed summarize biological disorders mechanisms.

Background: Periodontitis is an inflammatory condition initiated by oral bacteria and associated with several systemic diseases. Quercetin anti-inflammatory anti-bacterial poly-phenol present in various foods. The aim of this meta-analysis was the evaluation effects quercetin administration animal models experimental periodontitis. Methods: A systematic search performed electronic databases using following terms: “periodontitis” or “periodontal disease” “gingivitis” “quercetin” “cyanidanol” “sophoretin” “pentahydroxyflavone”. In vivo preclinical periodontal disease a measurement alveolar bone loss were included analysis. risk bias studies assessed SYRCLE tool. Results: yielded 335 results. Five included, four them qualified for meta-analysis. showed that decreased (τ2 = 0.31, 1.88 mm 95%CI: 1.09, 2.67) models. However, assessment indicated domains had high bias. Conclusions: diminishes prevents progression disease. might facilitate tissue hemostasis reducing senescent cells, decreasing oxidative stress via SIRT1-induced autophagy, limiting inflammation, fostering bacterial microenvironment symbiotic microbiota health. Future research will show whether how promising results can be translated into clinical treatment
