Several cardiovascular illnesses are associated with aberrant activation of cellular pyroptosis, ferroptosis, necroptosis, cuproptosis, disulfidptosis and macrophage polarisation as hallmarks contributing to vascular damage abnormal cardiac function. Meanwhile, these three novel forms dysfunction closely related mitochondrial homeostasis. Mitochondria the main organelles that supply energy maintain Mitochondrial stability is maintained through a series regulatory pathways, such fission, fusion mitophagy. Studies have shown (e.g., impaired dynamics mitophagy) promotes ROS production, leading oxidative stress, which induces M1 phenotypic polarisation. Therefore, an in-depth knowledge dynamic regulation mitochondria during necessary understand disease development. This paper systematically summarises impact changes in mitophagy on regulating dysfunctions promote understanding pathogenesis diseases provide corresponding theoretical references for treating diseases.

Cell death occurs continuously throughout individual development. By removing damaged or senescent cells, cell not only facilitates morphogenesis during the developmental process, but also contributes to maintaining homeostasis after birth. In addition, reduces spread of pathogens by eliminating infected cells. is categorized into two main forms: necrosis and programmed death. Programmed encompasses several types, including autophagy, pyroptosis, apoptosis, necroptosis, ferroptosis, PANoptosis. Autophagy, a mechanism that maintains cellular equilibrium via breakdown reutilization proteins organelles, implicated in regulating almost all forms pathological contexts. Notably, PANoptosis are directly classified as autophagy-mediated Therefore, autophagy presents therapeutic approach for treating diseases such inflammation tumors arise from abnormalities other This review focuses on crosstalk between modalities, providing new perspectives clinical interventions inflammatory neoplastic diseases.

The health-beneficial effects of nutraceuticals in various diseases have received enhanced attention recent years. Aging is a continuous process wherein physiological activity an individual declines over time and characterized by indefinite hallmarks which contribute toward aging-related comorbidities include many neurodegenerative diseases, cardiac problems, diabetes, bone-degeneration, cancer. Cellular senescence homeostatic biological that has important function driving aging. Currently, growing body evidence substantiates the connection between epigenetic modifications aging process, along with diseases. These are now being recognized as promising targets for emerging therapeutic interventions. Considering almost all processes modulated RNAs, numerous RNA-binding proteins been found to be linked age-related complexities. studies shed light on ability nutraceutical Withania somnifera (Ashwagandha) influence RNA expression, stability, processing, offering insights into its mechanisms action. By targeting RNA-related pathways, may exhibit ameliorating age-associated molecular changes, gene expression signaling networks. This review summarizes potential role modulating RNA-level changes associated aging, encompassing both vitro vivo studies. Taken together, putative role(s) modulation key RNAs will provide understanding facilitate development preventive strategies employing healthy

Diabetes mellitus (DM) is a metabolic illness defined by elevated blood glucose levels, mediating various tissue alterations, including the dysfunction of vital organs. can lead to many consequences that specifically affect brain, heart, and kidneys. These issues are known as neuropathy, cardiomyopathy, nephropathy, respectively. Inflammation acknowledged pivotal biological mechanism contributes development diabetes consequences. NF-κB modulates inflammation immune system at cellular level. Its abnormal regulation has been identified in several clinical situations, cancer, inflammatory bowel illnesses, cardiovascular diseases, Mellitus (DM). The purpose this review evaluate potential impact on complications associated with DM. Enhanced activity promotes inflammation, resulting harm compromised organ performance. Phytochemicals, which therapeutic molecules, potentially decline level, therefore alleviating progression problems correlated More importantly, be influenced factors, such TLR4 Highlighting these factors facilitate novel therapies future.

The constant increase in the elderly population presents significant challenges addressing new social, economic, and health problems concerning this population. With respect to health, aging is a primary risk factor for age-related diseases, which are driven by interconnected molecular hallmarks that influence development of these diseases. One main mechanisms has attracted more attention autophagy, catabolic process removes recycles damaged or dysfunctional cell components preserve viability. autophagy can be induced deregulated response wide range internal external stimuli, such as starvation, oxidative stress, hypoxia, organelles, infectious pathogens, aging. Natural compounds promote stimulation regulatory pathways, mTOR, FoxO1/3, AMPK, Sirt1, lead increased levels essential proteins Beclin-1 LC3, well decrease p62. These changes indicate activation autophagic flux, known decreased cardiovascular neurodegeneration, cataracts. regulated administration natural offers an adjuvant therapeutic alternative diseases; however, experimental evidence needed support confirm benefits. Hence, review aims highlight potential benefits regulating pathways approach combating

Abstract Coronary artery disease (CAD) poses a substantial threat to global health, leading significant morbidity and mortality worldwide. It has genetic component that been studied through genome-wide association studies (GWAS) over the past 17 years. These have made progress with larger sample sizes, diverse ancestral backgrounds, discovery of multiple genomic regions related CAD risk. In this review, we provide comprehensive overview GWAS, including information about makeup importance ethnic diversity in these studies. We also discuss challenges identifying causal genes variants within GWAS loci focus on non-coding regions. Additionally, highlight tissues cell types relevant CAD, clinical implications findings polygenic risk scores, sex-specific differences genetics, ethnical aspects personalized interventions, guided drug development.

Autophagy is a vital cellular pathway in eukaryotic cells, including neurons, where it plays significant roles neurodevelopment and maintenance. A crucial step autophagy the formation of class III phosphatidylinositol 3-kinase complex 1 (PI3KC3-C1), which essential for initiating autophagosome biogenesis. Beclin key component PI3KC3-C1, its interactors have been reported to affect autophagy. The brain-enriched adaptor protein FE65 has shown interact with Alzheimer’s disease amyloid precursor (APP) alter processing APP. Additionally, implicated various pathways, We demonstrate here that positively regulates FE65, through C-terminus, 1. Notably, overexpression enhances 1-mediated autophagy, whereas this process attenuated knockout cells. Moreover, stimulatory effect on diminished by an C-terminus deletion mutant disrupts FE65–Beclin interaction. Lastly, we found FE65-Beclin interaction modulates kinase activity PI3KC3-C1 complex. Together, identified as novel interactor, potentiates
