The gut microbiome plays a crucial role in human health by influencing various physiological functions through complex interactions with the endocrine system. These involve production of metabolites, signaling molecules, and direct communication cells, which modulate hormone secretion activity. As result, can exert neuroendocrine effects contribute to metabolic regulation, adiposity, appetite control. Additionally, influences reproductive altering levels sex hormones such as estrogen testosterone, potentially contributing conditions like polycystic ovary syndrome (PCOS) hypogonadism. This study aimed to assess the causal relationship between lipidome and female reproductive diseases (FRDs) using an advanced series of Mendelian randomization (MR) methods. utilized genome-wide association (GWAS) summary statistics encompassing 179 lipidomes six prevalent FRDs, namely polycystic ovary syndrome (PCOS), endometriosis, uterine fibroid, infertility, endometrial cancer, ovarian cancer. The two-sample MR (TSMR) approach was employed investigate relationships, with further validation false discovery rate (FDR) multivariable (MVMR) Subsequently, a range comprehensive evaluations were performed, including sensitivity analysis, mediation reverse steiger test. Examining traits as exposures 6 FRDs outcomes, this identified significant effects 56 lipids on FRDs. Following multiple testing correction MVMR validation, sphingomyelin (d38:2) found have protective effect against PCOS (β = -0.104, 95% CI: -0.199 ~ -0.010, P 0.031). Phosphatidylcholine (18:0_22:6) associated decreased risk developing fibroid -0.111, -0.201~ -0.021, 0.016), sterol ester (27:1/20:3) showed significance in cancer -0.248, -0.443 -0.053, 0.013). Conversely, phosphatidylethanolamine (18:2_0:0) increased endometriosis 0.183, 0.015 0.350, 0.033), while (27:1/18:1) posed influence 1.007, 0.925 1.089, < 0.001), phosphatidylcholine (16:0_22:6) 0.229, 0.039 0.420, 0.018). Furthermore, it determined that associations these profiles independent BMI, obesity, diabetes, smoking, alcohol use, physical activity, inflammation, depression, waist-hip ratio, vitamin D, dehydroepiandrosterone sulphate, sex hormone binding globulin, testosterone levels. Most outcomes passed consistent tests without evidence heterogeneity, pleiotropy, or causality. results indicated close specific lipidomes, particularly sphingomyelin, lysophosphatidylethanolamine, cholesterol ester, phosphatidylcholines, These lipid species may potentially serve biomarkers future drug targets for treatment

We developed a prediction model for postprandial glycemic response (PPGR) in pregnant women, including those with diet-treated gestational diabetes mellitus (GDM) and healthy explored the role of gut microbiota improving accuracy. The study involved 105 women (77 GDM, 28 healthy), who underwent continuous glucose monitoring (CGM) 7 days, provided food diaries, gave stool samples microbiome analysis. Machine learning models were created using CGM data, meal content, lifestyle factors, biochemical parameters, data (16S rRNA gene sequence analysis). Adding increased explained variance peak levels (GLUmax) from 34 to 42% incremental area under curve (iAUC120) 50 52%. final showed better correlation measured PPGRs than one based only on carbohydrate count (r = 0.72 vs. r 0.51 iAUC120). Although features important, their contribution performance was modest.

Previous studies suggested that immune factors may play critical roles in female infertility, but their causal links remain unclear. To address this gap, study employs the Mendelian randomization (MR) to delineate association between circulating and infertility. This employed summary-level data from three genome-wide (GWAS) encompassing 731 peripheral cell signatures, 41 cytokines, five infertility phenotypes reveal relationship Causalities of exposure-outcome pairs were explored mainly using two-sample MR, comprehensive sensitivity analyses deployed validate reliability results. Multi-variable (MVMR) was further examine potential mediating effects significant exposures. Following false discovery rate (FDR) correction analyses, univariable identified distinct signatures across subtypes. Peripheral levels Naive CD8br %CD8br, MIP1B IL17 causally associated with general higher level decreased risk ovarian Furthermore, CD80 on monocyte a tubal features (CD86 + myeloid DC AC, HLA DR NK %NK, CD16 CD14- monocyte) for uterine factor cytokines (MIP1B, IL18, IL17) genetic causes cervical vaginal other or unspecified origin (FIOTHNAS). MVMR revealed MIP1B's FIOTHNAS substantially attenuated upon adjusting IL18. Our results highlight response contributes through subtype-specific mechanisms, providing clues following clinical research treatment.

Previous studies have reported that alterations in gut microbiota composition are associated with male fertility. However, it is unclear and difficult to establish whether these associations reflect a causal relationship. We assessed genome-wide association study summary statistics for fertility perform MR analysis. Independent single nucleotide polymorphisms closely 211 bacterial taxa (N = 122,110) were identified as instrumental variables. The statistic data infertility 733,479), abnormal spermatozoa 209,921) erectile dysfunction 223,805) obtained from the latest release FinnGen consortium outcome of interest. Two-sample was performed evaluate effect on fertility, including inverse-variance-weighted (IVW) method, weighted median MR-Egger, mode-based estimation MR-PRESSO. It aimed to explore potential cause-and-effect relationship bacteria problems such as inflammation, infertility, sperm functionality, using a two-sample Mendelian randomization method examine these connections. analysis found certain bacterial genera, Erysipelatoclostridium (0.71 [0.55–0.92]), Parasutterella (0.74 [0.57–0.96]), Ruminococcaceae UCG-009 (0.77 [0.60–0.98]), Slackia (0.69 [0.49–0.96]), showed protective effects against prostatitis. In contrast, other genera like Faecalibacterium (1.59 [1.08–2.34]), Lachnospiraceae UCG004 (1.64 [1.15–2.34]), Odoribacter (1.68 [1.01–2.81]), Paraprevotella (1.28 [1.03–1.60]), Sutterella (1.58 [1.13–2.19]) detrimental. Additionally, identified 2 orchitis epididymitis, 3 5 abnormal spermatozoa. Further of sperm-related proteins revealed associations specific SPACA3, SPACA7, SPAG11A, SPAG11B, SPATA9, SPATA20, ZPBP4. results remained robust after sensitivity reverse analysis. concluded have roles in sperm-associated proteins.
