Coronavirus disease 2019 (COVID-19) is currently a worldwide emergency caused by Severe Acute Respiratory Syndrome 2 (SARS-CoV-2). In observational clinical studies, statins have been identified as beneficial to hospitalized patients with COVID-19. However, experimental evidence of underlying protection against SARS-CoV-2 remains elusive. Here we reported for the first-time protective effects simvastatin treatment both in vitro and vivo . We found that significantly reduced viral replication lung damage , delaying SARS-CoV-2-associated physiopathology mortality K18-hACE2-transgenic mice model. Abstract The coronavirus disease 2019 (COVID-19) caused by SARS-CoV-2 infection has become a global pandemic due to the high viral transmissibility and pathogenesis, bringing enormous burden our society. The rapid development of vaccines to combat severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections has been critical reduce the severity COVID-19. The spread of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and its variants is threatening public health around the world. Endocytosis functions as an important way for viral infection, SARS-CoV-2 bears no exception. However, specific endocytic mechanism remains unknown. In this study, we used inhibitors to evaluate role different routes in pseudovirus infection found that was associated with caveolar/lipid raft- cytoskeleton-mediated endocytosis, but independent clathrin-mediated endocytosis macropinocytosis. Meanwhile, knockdown CD147 Rab5a Vero E6 Huh-7 cells inhibited co-localization spike protein, CD147, observed pseudovirus-infected cells, which weakened by silencing, illustrating entered host via CD147-mediated endocytosis. Additionally, Arf6 silencing markedly while little change knockout-Vero cells. This finding indicated Arf6-mediated trafficking plays a vital entry. Taken together, our findings provide new insights into CD147-Arf6 axis mediating entry further suggest blockade pathway seems be feasible approach prevent clinically.

BackgroundHost cell-membrane cholesterol, an important player in viral infections, is constant interaction with serum high-density lipoprotein-cholesterol (HDL-C) and low-density (LDL-C). Low lipid levels during hospital admission are associated COVID-19 severity. However, the effect of antecedent on SARS-CoV-2 infection risk has not been explored.MethodsFrom our retrospective cohort from Arkansas Clinical Data-Repository, we used log-binomial regression to assess among trajectories 2 years testing, identified using group-based-trajectory modelling. We mixed-effects linear level trends followed up time of, 2-months following testing.FindingsAmong 11001 individuals a median age 59 (IQR 46-70), 1340 (12.2%) tested positive for COVID-19. The highest trajectory HDL-C was lowest (RR 0.63, 95%CI 0.46-0.86). Antecedent LDL-C, total cholesterol (TC), triglycerides (TG) were independently risk. In patients, (-7.7, -9.8 -5.5 mg/dL), LDL-C (-6.29, -12.2 -0.37 but TG levels, decreased transiently at testing.InterpretationHigher HDL-C, TC, or TG, lower Serum declined infection.
