Abstract Cytokine storm (CS) is a severe systemic inflammatory syndrome characterized by the excessive activation of immune cells and significant increase in circulating levels cytokines. This pathological process implicated development life-threatening conditions such as fulminant myocarditis (FM), acute respiratory distress (ARDS), primary or secondary hemophagocytic lymphohistiocytosis (HLH), cytokine release (CRS) associated with chimeric antigen receptor-modified T (CAR-T) therapy, grade III to IV graft-versus-host disease following allogeneic hematopoietic stem cell transplantation. The involvement JAK-STAT pathway, Toll-like receptors, neutrophil extracellular traps, NLRP3 inflammasome, other signaling pathways has been recognized pathogenesis CS. Therapies targeting these have developed are currently being investigated. While novel drugs demonstrated promising therapeutic efficacy mitigating CS, overall mortality rate CS resulting from underlying diseases remains high. In clinical setting, management typically necessitates multidisciplinary team strategy encompassing removal abnormal system activation, preservation vital organ function, treatment disease, provision life supportive therapy. review provides comprehensive overview key cytokines elucidates impact dysregulated delineates resultant injury addition, we offer insights current literature on cases FM, ARDS, response syndrome, treatment-induced CRS, HLH, related conditions.

Abstract Traditionally, lactate has been considered a ‘waste product’ of cellular metabolism. Recent findings have shown that is substance plays an indispensable role in various physiological functions and contributes to energy metabolism signal transduction during immune inflammatory responses. The discovery lactylation further revealed the regulating processes. In this review, we comprehensively summarize paradoxical characteristics microenvironment highlight pivotal roles homeostasis, shuttle, (‘lactate clock’) acute chronic responses from molecular perspective. We especially focused on receptors with either proinflammatory or anti-inflammatory effects complex biological signalling pathways investigated dynamic changes cells lactate-related microenvironment. Moreover, reviewed progress use as therapeutic target for response, which may provide new perspective treating inflammation-related diseases.

Chronic Human Immunodeficiency Virus (HIV) infection remains a significant challenge to global public health. Despite advances in antiretroviral therapy (ART), which has transformed HIV from fatal disease into manageable chronic condition, definitive cure elusive. One of the key features is immune activation and inflammation, are strongly associated with, predictive of, progression, even patients successfully treated with suppressive ART. inflammation characterized by persistent cell metabolic dysregulation, cellular exhaustion dysfunction. This review aims summarize current knowledge interplay between metabolism, T dysfunction infection, also discusses use humanized mice models study pathogenesis develop novel therapeutic strategies.

There is evidence that the triazine herbicide atrazine, which used extensively, present in both surface water and groundwater, its interfering effect on immune systems, endocrine tumours has been reported by laboratory epidemiological studies. This study explored how atrazine affected 4T1 breast cancer cell development vitro vivo. The obtained results showed after exposure to proliferation tumour volume were significantly increased expression of MMP2, MMP7, MMP9 was upregulated. thymus spleen indices, CD4 + CD3 lymphocyte percentages from inguinal lymph nodes, /CD8 ratio noticeably lower than they control group. Importantly, tumour-infiltrating lymphocytes such as , CD8 NK cells decreased while Treg increased. Moreover, IL-4 IFN-γ TNF-α serum microenvironment. Autoimmune diseases are typically characterized by aberrant activation of immune system that leads to excessive inflammatory reactions and tissue damage. Nevertheless, precise targeted efficient therapies limited. Thus, studies into novel therapeutic targets for the management autoimmune urgently needed. Radical S-adenosyl methionine domain-containing 2 (RSAD2) is an interferon-stimulated gene (ISG) renowned antiviral properties protein it encodes, named viperin. An increasing number have underscored new roles RSAD2/viperin in immunomodulation mitochondrial metabolism. Previous shown there a complex interplay between RSAD2/vipeirn mitochondria binding iron-sulfur (Fe-S) cluster necessary involvement viperin Viperin influences proliferation development cells as well inflammation via different signaling pathways. However, function varies comprehensive overview this emerging theme lacking. This review will describe characteristics RSAD2/viperin, decipher its immunometabolic processes, clarify crosstalk mitochondria. Furthermore, we emphasize crucial RSAD2 potential application value.
