Abstract The 5-year survival rate of patients with head and neck squamous cell carcinoma (HNSCC) remains <50% due to risk the recurrence metastasis. Hypoxia patterns are a hallmark HNSCC that associated its occurrence progression. However, precise role hypoxia during HNSCC, such as relationship between hypoxia, tumor immune landscape communication orchestration largely unknown. current study integrated data from bulk single-cell RNA sequencing analyses define HNSCC. First, scoring system named score (HS) was constructed based on hypoxia-related genes (HRGs) expression. outcomes, stromal/immune/Estimate scores, responses targeted inhibitors, chemotherapeutic, immunotherapy were distinct high HS group low group. Single-cell datasets showed different distributions in populations. Furthermore, HLA-DPA1/CD4 axis identified unique interaction CD4+ T Conv pDC cells. These findings may enhance understanding characteristics

Metabolic plasticity and ferroptosis are essential for colorectal cancer (CRC) progression. The effects prognostic value of metabolic plasticity- ferroptosis-related genes (MPFRGs) in CRC remain unclear. We established a model patients by identifying important ferroptosis. Data were retrieved from Cancer Genome Atlas (TCGA) Gene Expression Omnibus; MPFRG data obtained GeneCards FerrDb. performed functional (to explore differences between the two subtypes) single-sample gene set assess immune environment) enrichment analyses. Immunophenotype, tumor immunological dysfunction, exclusion scores assessed to determine patient responses. A least absolute shrinkage selection operator-Cox regression comprising 10 significant differentially expressed (MPFDEGs) was constructed using TCGA training cohort validated GSE17536 GSE39582 datasets. nomogram ferroptosis-based signatures, revealing clinical application potential molecular mechanisms underlying role MPFRGs CRC. Our (developed based on MPFDEGs) is efficient calculating overall survival patients. findings provide new strategies management individualized treatment these

Background: Calcium signaling is implicated in multiple processes including immune response that important tumor progression. Kidney renal clear cell carcinoma (KIRC) the most frequent histological type of with up to a third cases develop metastases. As result lack in-depth understanding mechanisms underlying KIRC, treatment options have been limited. Here, we aim comprehensively investigate landscape Ca 2+ channels, pumps and exchangers KIRC patients. Methods: The mRNA expression profiles gene variations 58 calcium-related genes (CRGs) patients normal control were downloaded from TCGA database. CRGs-related risk score was constructed quantify calcium patterns by using least absolute shrinkage selection operator (LASSO) regression. prognostic value, biological functions, therapeutic sensitivities based on then evaluated methods. Finally, key CRGs identified weighted co-expression network analysis (WGCNA). TCGA-CPTAC, GSE53757 datasets, as well human tissues used for validation. Results: had significant differences CRG expression, prognosis, functions between two clusters. determined. mutation burden, infiltration, checkpoints, targeted inhibitors remarkably different high low subtypes. high-risk subtype characterized immunosuppressive microenvironment poor prognosis. Meanwhile, several drugs showed distinct sensitivity TRPM3 protein significantly lower tumors than controls. Low associated prognosis Conclusion: Our study highlighted promising value tumors. evaluation will contribute predicting clinical therapy

Background: Pancreatic adenocarcinoma (PAAD) is a highly malignant and aggressive tumor with high incidence, mortality, poor prognosis. Exploring the molecular features associated PAAD development will help to improve patient treatment PANoptosis new form of cell death that variety diseases. However, its mechanism in are unclear. Therefore, exploring role clinical significance urgently needed. Methods: Based on pertinent information from The Cancer Genome Atlas (TCGA), Gene Expression Omnibus (GEO), Genotype-Tissue (GTEx) databases, we examined function PANoptosis-related genes (PRGs) screened 146 differentially expressed (PRDEGs). prognostic, therapeutic, subtype categorization potential PRDEGs patients were examined. Results: Unsupervised clustering was performed using consistent algorithm, which could classify into three groups based PRDEGs. PRDEGs, constructed risk assessment model containing five genes: TNNT1, SEZ6L, S100A14, CXCL10, MATN3. Using this model, can serve as standalone prognostic indicator, divided high- low-risk groups. abnormal expression these verified bioinformatic analysis experiments. Conclusions: In summary, study discovered plays vital regulatory developed predict PAAD, offer novel theoretical framework for management PAAD.

Abstract Background Small bowel cancer (SBC) is a very rare solid malignancy. Consequently, compared with other malignant gastrointestinal tumors, our knowledge regarding SBC, specifically its molecular attributes, remains limited. Herein, we aim to provide an overview of the gene characteristics Chinese patients We particularly focus on elucidating genetic intricacies that differentiate SBC whose primary tumors originate in distinct anatomical regions within small bowel. Methods During period ranging from February 2018 December 2022, total 298 tumor samples were consecutively collected diagnosed cancer.. Next-generation sequencing (NGS) was performed detect mutation, assess microsatellite instability (MSI), and evaluate mutational burden (TMB). Additionally,, IHC used analyze level PD-L1 expression samples. Results The outcomes next-generation unveiled predominant mutations observed (SBC). top ten identified as follows: TP53 (53%), KRAS (51%), APC (31%), SMAD4 (19%), VEGFA (15%), CDKN2A RAC1 LRP1B (14%), MGMT (14%, CD74 (13%). Subsequent analysis revealed disparities landscape between cohort this study Memorial Sloan Kettering Cancer Center (MSKCC), Notably, distinguishable frequencies several genes, including ERBB2, FBXW7, PIK3CA, etc . which exhibited contrasting presence both MSKCC cohort.. Furthermore, noticed variations frequency among depending specific site where originated In addition, distribution high (MSI-H) (TMB) levels varied originating duodenum, jejunum, ileum. Conclusion profile comparison populations, highlighting unique landscape. noticeable situated duodenum those affecting bowel, suggests these should be treated differently.
