Rheumatoid arthritis (RA) is a common autoimmune disease characterized primarily by persistent synovial inflammation and joint destruction. In recent years, ferroptosis, as novel form of cell death, has garnered widespread attention due to its critical role in various diseases. This review explores the potential mechanisms ferroptosis RA relationship with pathogenesis RA, systematically analyzing regulatory cells, chondrocytes, immune cells. We emphasize evaluation ferroptosis-related pathways their therapeutic targets, including development application inhibitors activators. Although shows some promise treatment, dual safety issues clinical still require in-depth study. Future research should focus on elucidating specific pathology developing more effective safer strategies provide new treatment options for patients.

Iron is essential in various physiological processes, but its accumulation leads to oxidative stress and cell damage, thus iron homeostasis has be tightly regulated. Ferroptosis an iron-dependent non-apoptotic regulated death characterized by overload ROS accumulation. Mitochondria are organelles playing a crucial role metabolism involved ferroptosis. MitoNEET, protein of mitochondrial outer membrane, key element this process. Ferroptosis, altering levels several metabolically active organs, linked non-communicable diseases. For example, the liver hepatic fibrosis cirrhosis, accelerating NAFLD progression, muscle cells contributes damage leading sarcopenia, brain associated neurodegeneration. The aim review investigate intricate balance regulation focusing on mitochondria stress, analyzing ferroptosis implications health disease.

With the global population aging, musculoskeletal disorders (MSDs) have posed significant physical and psychological health challenges for patients as well a substantial economic burden on society. The advancements in conservative surgical interventions MSDs been remarkable recent years; however, current treatment modalities still fall short of meeting optimal requirements patients. Recently, peroxiredoxin 6 (Prdx6) has gained considerable attention from researchers due to its antioxidative, anti-inflammatory, anti-apoptotic properties. It found that Prdx6 is involved multiple system diseases, including MSDs; exact role lacking. This study aimed summarize structure, regulatory mechanism, potential function Prdx6. These findings may demonstrate novel target inhibiting advancement MSDs.

Abstract Cadmium (Cd) and excess molybdenum (Mo) are multiorgan toxic, but the detrimental impacts of Cd and/or Mo on poultry have not been fully clarified. Thence, a 16‐week sub‐chronic toxic experiment was executed with ducks to assess toxicity Mo. Our data substantiated that coexposure evidently reduced GSH‐Px, GSH, T‐SOD, CAT activities elevated H 2 O MDA concentrations in myocardium. What is more, study suggested united exposure synergistically Fe 2+ content myocardium activated AMPK/mTOR axis, then induced ferroptosis by obviously upregulating ACSL4, PTGS2, TFRC expression levels downregulating SLC7A11, GPX4, FPN1, FTL1, FTH1 levels. Additionally, further caused excessive ferritinophagy observably increasing autophagosomes, colocalization endogenous LC3, ATG5, ATG7, LC3II/LC3I, NCOA4, In brief, this for first time finally augmenting injure ducks, which will offer an additional view between two heavy metals poultry.

Abstract Context With the development of society, number patients with osteoporosis is increasing. The prevention and control has become a serious urgent issue. continuous progress biomedical research, ferroptosis attracted increased attention. However, pathophysiology mechanisms still need further study. Natural products are widely used in East Asian countries for treatment. Objective In this paper, we will discuss basic ferroptosis, relationship between osteoclasts osteoblasts, vitro vivo studies natural to prevent by interfering ferroptosis. Methods This article takes products, osteoporosis, osteoblasts osteoclast as key words. Retrieve literature from 2012 2023 indexed databases such PubMed Central, PubMed, Web Science, Scopus ISI. Results Ferroptosis many regulatory mechanisms, including system XC -/GSH/GPX4, p62/Keap1/Nrf2, FSP1/NAD (P) H/CoQ10, P53/SAT1/ALOX15 axes etc. Interestingly, found that Artemisinin, Biochanin A Quercetin, can play role treating promoting inhibiting osteoblasts. Abstract Iron homeostasis is crucial for maintaining proper cellular function, and its disruption considered one of the pathogenic mechanisms underlying musculoskeletal diseases. Under conditions oxidative stress, accumulation iron overload lipid peroxidation can lead to ferroptosis. Extracellular vesicles (EVs), serving as mediators in cell‐to‐cell communication, play an important role regulating outcome cell Growing evidence has proven that EV biogenesis secretion are tightly associated with export. Furthermore, different sources EVs deliver diverse cargoes bring about phenotypic changes recipient cells, either activating or inhibiting Thus, delivering therapies targeting ferroptosis through may hold significant potential treating This review aims summarize current knowledge on ferroptosis, well their therapeutic applications diseases, thereby provide valuable insights both research clinical practice.

Osteoarthritis (OA) is the most common chronic joint disease and how ferroptosis contributes to OA has garnered much attention recently. Bioinformatics promoted discovery of ferroptosis-related biomarkers for OA. But since a whole-joint disease, sensitive are still limited. We herein focused on subchondral bone, component often-ignored by existing bioinformatic reports, identify diagnostic OA.Microarray datasets GSE51588 GSE55457 were downloaded from Gene Expression Omnibus database. Ferroptosis-related differential expression genes (Ferr-DEGs) between normal samples identified their functional enrichment was analyzed. Common diagnosis selected Ferr-DEGs using combination SVM-RFE, LASSO regression, RandomForest machine learning algorithms. genes' value verified receiver operating characteristic (ROC) curve association with immune infiltration analyzed CIBERSORT. Finally, candidate gene's in chondrocytes patients an vitro IL-1β-induced model, RT-PCR.Two genes, LPCAT3 PGD, as confirmed ROC test. The PGD several types cells identified. decreased both condition. We potential OA, which may offer insight into role provides useful information treatment
