Rheumatoid arthritis (RA) is a chronic immunocompromised disorder that primarily affects joints thereby leading to synovial inflammation, pain, stiffness, stress, and affecting the quality of life. The present study aimed determine identify bioactive compounds can be developed as potential inhibitors target human TNF-α IL-6 proteins associated with RA. also physiochemical properties toxicity by ADMET, binding pockets via CASTp server, stereochemical protein PROCHECK, MMGB/SA analysis fastDRH molecular dynamics (WebGro). findings revealed staurosporine was most effective compound against highest affinity -10.8 kcal/mol. Similarly, withanoside L showed -8.2 kcal/mol when docked IL-6. In this study an in situ forming gel for curcumin and piperine delivery is investigated as a long-lasting strategy the local treatment of inflammatory degenerative joint disease, such osteoarthritis rheumatoid arthritis. Qingre Huoxue Decoction (QRHXD) is a traditional Chinese herbal prescription widely used in clinical practice with significant therapeutic effects on RA; however, its mechanism of action remains unclear. This study aimed to investigate the efficacy and underlying mechanisms QRHXD treating RA through research, multiomics approaches, animal experiments. We conducted 24-week which was primary treatment, collecting serum samples from patients before after treatment for integrated proteomic metabolomic analysis identify potential targets. Bioinformatics differentially expressed proteins (DEPs) differential metabolites (DMs) performed using hierarchical clustering, volcano plots, heat maps, Gene Ontology (GO), Kyoto Encyclopaedia Genes Genomes (KEGG) analysis. To validate identified targets, we constructed collagen-induced arthritis (CIA) mouse model. Clinical research has shown that can improve symptoms relevant indicators patients, including disease activity score-28 (DAS28), C-reactive protein (CRP), erythrocyte sedimentation rate (ESR), tender joint count (TJC), swollen (SJC), visual analogue scale (VAS), patient-reported outcome (PRO), health assessment questionnaire (HAQ). Proteomics metabolomics 83 DEPs 54 DMs, 46 upregulated 37 downregulated proteins, as well 11 43 metabolites. KEGG enrichment revealed are primarily associated fatty acid degradation, ferroptosis, glycerolipid metabolism, related pathways. The DMs AMPK signalling pathway, FoxO glycolysis/gluconeogenesis, MTOR so on. Rheumatoid Arthritis (RA), an autoimmune disease, is a chronic inflammatory disorder affecting the joints leading to severe damage and cartilage destruction. ABSTRACT Periodontitis (PDS) is one of the most common and crippling systemic diseases. It a chronic inflammatory condition that leads to loss periodontal attachment, resulting in tooth loss. In addition its effects on oral health nutrition, PDS closely linked other conditions such as diabetes mellitus (DM), cardiovascular diseases (CVD), rheumatoid arthritis (RA). Introduction Inflammatory Bowel Disease (IBD), encompassing Crohn disease (CD) and ulcerative colitis (UC), presents complex challenges in management due to dysregulated immune responses genetic predispositions. This study explored the potential of curcumin as an adjunctive therapy IBD, assessing its efficacy safety through a systematic review clinical trials enhance treatment strategies outcomes. Methods To identify placebo-controlled randomized on databases such Medline/PubMed, Scopus, Embase, Web Knowledge, Google Scholar were searched till May 2024. Inclusion criteria focused RCTs comparing with placebo IBD patients, data extraction analysis conducted using established methodologies tools for comprehensive synthesis assessment findings. Results In this meta-analysis, 13 included after screening 362 records conducting full-text review. Most UC patients published post-2010, utilizing oral varying dosages durations. The showed curcumin’s significant achieving remission response heterogeneity observed. Adverse events withdrawal rates did not significantly differ between groups. CD show superiority over endoscopic remission. Conclusion findings highlight but indicate inconclusive results CD, emphasizing need further research. multifaceted mechanisms involve anti-inflammatory, antioxidant, microbiota modulatory, immune-regulating properties. Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent inflammation and progressive joint destruction. Recent data underscore oxidative stress as primary factor in the pathophysiology of rheumatoid arthritis, intensifying inflammatory processes tissue damage via overproduction reactive oxygen species (ROS) compromised antioxidant defenses. Current therapies, including disease-modifying antirheumatic drugs (DMARDs), primarily target immune dysregulation but fail to address stress, necessitating novel adjunctive treatment strategies. This review explores potential antioxidant-based therapies complementary approaches RA management. Natural compounds such curcumin, resveratrol, sulforaphane, propolis exhibit strong anti-inflammatory antioxidative properties modulating redox-sensitive pathways, nuclear kappa-light-chain-enhancer activated B cells (NF-κB) erythroid 2-related 2(Nrf2)/heme oxygenase (HO-1). N-acetylcysteine (NAC) replenishes intracellular glutathione, enhancing cellular resilience against stress. Additionally, molecular hydrogen (H2) selectively neutralizes harmful ROS, reducing inflammation.
