Sjögren's syndrome (SS) is a systemic autoimmune disease, with major symptoms including dry mouth and eyes, for which there no effective treatment. Recent studies have demonstrated that mesenchymal stem cells (MSCs) are in the treatment of SS, but efficacy allogeneic MSCs affected by variability among different cell donors, they easily cleared immune system recipient. Autologous one ideal options SS; however, their function decreases age. Regenerative fibroblast (rFib) type new MSC obtained through chemical reprogramming technology from skin fibroblasts. In this study, we report safety intravenous infusion autologous rFib volunteer SS. March 2021, was diagnosed SS due to positive anti-SSB antibodies, lymphocyte infiltration lip gland, large area purpura both lower limbs. From May 2021 November 2022, she received Umbilical cord (UCMSC) therapy (5.0 × 107 UCMSCs per time, totaling 10 infusions), her condition did not improve. 2023, prepared, meeting quality standard T/CSCB0003-2021 Human Mesenchymal Stem Cells. Between October 2023 June 2024, total 12 transfusions rFib. After treatments, experienced recurrence Symptoms mouth, fatigue were relieved. ESR, B lymphocytes, rheumatoid factor IgM, IgA declined, while proportion NK increased, most cytokines returned normal levels. vitro experiments showed could significantly inhibit proliferation T lymphocytes after PHA stimulation. No adverse effects associated use during clinical trial. The results trial indicate injections safe treating may be more suitable diseases than MSCs.

Type I interferon (IFN-I) plays crucial roles in the regulation of inflammation and it is associated with various inflammatory diseases including systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), periodontitis, impacting people's health quality life. It well-established that IFN-Is affect immune responses factors by regulating some signaling. However, currently, there no comprehensive overview regulatory role IFN-I distinctive pathways as well diseases. This review aims to provide a narrative involvement different signaling pathways, mainly mediating related key specific targets cascades influence progression As such, we suggested induce through stimulation certain which displays possible efficient treatment methods provides reference for precise control

Inflammasome complexes and their integral receptor proteins have essential roles in regulating the innate immune response inflammation at post-translational level. Yet despite protective role, aberrant activation of inflammasome gain function mutations component genes seem to contribute development progression human autoimmune autoinflammatory diseases. In past decade, our understanding biology mechanisms has greatly progressed. We therefore provide an up-to-date overview various inflammasomes known action. addition, we highlight involvement pathogenic common autoinflammatory, neurodegenerative diseases, including atherosclerosis, rheumatoid arthritis, systemic lupus erythematosus, inflammatory bowel disease, Alzheimer’s Parkinson’s multiple sclerosis. conclude by speculating on future avenues research needed better understand health disease.

Abstract Sex characteristics exhibit significant disparities in various human diseases, including prevalent cardiovascular cancers, metabolic disorders, autoimmune and neurodegenerative diseases. Risk profiles pathological manifestations of these diseases notable variations between sexes. The underlying reasons for sex encompass multifactorial elements, such as physiology, genetics, environment. Recent studies have shown that body systems demonstrate sex-specific gene expression during critical developmental stages editing processes. These genes, differentially expressed based on different sex, may be regulated by androgen or estrogen-responsive thereby influencing the incidence presentation cardiovascular, oncological, metabolic, immune, neurological across However, despite existence differences patients with treatment guidelines predominantly rely male data due to underrepresentation women clinical trials. At present, there exists a substantial knowledge gap concerning mechanisms treatments diverse Therefore, this review aims elucidate advances examining epidemiological factors, pathogenesis, innovative progress accordance distinctive risk each disease provide new theoretical practical basis further optimizing individualized improving patient prognosis.

Abstract Background For the unclear pathogenesis of Sjogren's syndrome (SS), further exploration is necessary. Mesenchymal stem cells (MSCs) and derived exosomes (MSCs‐exo) have exhibited promising results in treating SS. Object This study aimed to investigate effect mechanism human umbilical cord MSCs (UC‐MSCs) on Methods Nonobese Diabetic (NOD) mouse splenic T were co‐cultured with UC‐MSCs UC‐MSCs‐exo, interferon‐gamma (IFN‐γ), interleukin (IL)‐6, IL‐10, prostaglandin E2 (PGE2), transforming growth factor‐β1 (TGF‐β1) levels supernatant assessed by quantitative real‐time polymerase chain reaction enzyme‐linked immunosorbent assay. Co‐cultured injected into NOD mice via tail vein. The inflammatory cell infiltration intestine submandibular gland was characterized hematoxylin‐eosin staining. Treg/Th17 homeostasis within spleen determined flow cytometry. Gut microbiota detected 16S rRNA sequencing, relationship between differential cytokines analyzed Pearson correlation coefficient. Results UC‐MSCs, successfully cultured identified. After both IFN‐γ IL‐6 decreased while PGE2, TGF‐β1 increased transcriptional translational levels. UC‐MSCs‐exo partially restored salivary secretion function, reduced Ro/SSA antibody α‐Fodrin immunoglobulin A levels, gland, raised proportion Treg cells, IFN‐γ, IL‐6, IL‐2, IL‐17, lipopolysaccharide, tumor necrosis factor‐alpha Foxp3, affecting cells. intervention improved intestinal increasing diversity richness. Additionally, significantly associated cytokine Conclusion Human disease characterization SS through regulation gut cellular immunity.

Abstract Autoimmune diseases commonly affect various systems, but their etiology and pathogenesis remain unclear. Currently, increasing research has highlighted the role of ferroptosis in immune regulation, with cells being a crucial component body’s system. This review provides an overview discusses relationship between ferroptosis, programmed cell death cells, autoimmune diseases. Additionally, it summarizes key targets such as GPX4 TFR, responses. Furthermore, release multiple molecules, including damage-associated molecular patterns (DAMPs), following by is examined, these molecules further influence differentiation function thereby affecting occurrence progression Moreover, secrete factors or metabolites, which also impact target organs tissues involved Iron chelators, chloroquine its derivatives, antioxidants, calreticulin have been demonstrated to be effective animal studies for certain diseases, exerting anti-inflammatory immunomodulatory effects. Finally, brief summary future perspectives on are provided, aiming guide disease treatment strategies.
