Abstract Gliomas are the most common primary neuroepithelial tumors of central nervous system in adults, which glioblastoma is deadliest subtype. Apart from intrinsically indestructible characteristics glioma (stem) cells, accumulating evidence suggests that tumor microenvironment also plays a vital role refractoriness glioblastoma. The functions platelets to stop bleeding and regulate thrombosis under physiological conditions. Furthermore, active elements participate variety processes development, including growth, invasion, chemoresistance. Glioma cells recruit activate resting become tumor-educated (TEPs), turn can promote proliferation, stemness, chemoresistance cells. TEPs be used obtain genetic information about gliomas, helpful for early diagnosis monitoring therapeutic effects. Platelet membranes intriguing biomimetic materials developing efficacious drug carriers enhance antiglioma activity. Herein, we review recent research referring contribution malignant gliomas focusing on molecular mechanisms mediating interaction between as well present challenges opportunities targeting therapy.

Fucoxanthin (FX) is a carotenoid of marine origin primarily distributed in brown seaweeds and has garnered interest for its antioxidative, anti-inflammatory, anticancer properties. Despite potential, comprehensive understanding effects mechanisms action remains elusive. The aim this review to present novel insights into the FX, shedding light on previously unexplored molecular synergistic potential with established chemotherapeutic agents. A search was conducted employing databases like PubMed/MedLine, Scopus, Web Science aggregate relevant pharmacological experimental studies. results studies showed that FX exhibits activity against various cancer types, including breast, colorectal, lung cancer, through multiple pathways: cell cycle arrest, apoptosis induction, inhibition angiogenesis. Additionally, potentiates existing agents, making it candidate combination therapies. evidence suggests possesses considerable properties, acting diverse mechanisms; heterogeneity study designs limited number clinical trials make hard conclude. Further in-depth studies, particularly randomized controlled trials, are essential validating FX's efficacy paving way integration standard treatment regimens; additional research needed explore pharmacokinetics, safety profile, chemotherapeutics.

Introduction: Vasculogenic mimicry (VM) represents a novel form of tumor angiogenesis that is associated with invasiveness and drug resistance. However, the VM landscape across cancer types remains poorly understood. In this study, we elucidate characterizations cancers based on multi-omics data provide potential targeted therapeutic strategies. Methods: Multi-omics from The Cancer Genome Atlas was used to conduct comprehensive analyses characteristics related genes (VRGs) types. Pan-cancer vasculogenic score established depiction correlation between phenotypes conducted explore regulatory mechanisms VM. We further systematically examined relationship both immunity microenvironment (TME). addition, cell communication analysis single-cell transcriptome investigate interactions cells TME. Finally, transcriptional response Genomics Drug Sensitivity in database were utilized identify targets drugs. impact immunotherapy also clarified. Results: Our study revealed VRGs dysregulated regulated by multiple mechanisms. Then, level found be heterogeneous among different tumors correlated invasiveness, metastatic potential, malignancy, prognosis. strongly epithelial-mesenchymal transition (EMT). Further cancer-associated fibroblasts can promote EMT formation. Furthermore, immune-suppressive state characterized high levels as an indicator predict effect immunotherapy. seven drugs targeting identified. Conclusion: conclusion, key various types, underscoring pivotal role CAFs immunosuppressive clues for research provides initial overview reference point future VM, opening up new avenues intervention.

The interplay between vascularization and macrophage-induced immune suppression plays a crucial role in melanoma treatment. In this study, we propose novel combination approach to combat by simultaneously inhibiting tumor enhancing macrophage-mediated anti-tumor responses. We investigate the potential of combining combretastatin A4 (CA4), vascular-disrupting agent, with poly(I:C) (PIC), an immunostimulatory adjuvant. This effectively suppresses cell proliferation, disrupts vascularization, promotes macrophage polarization towards M1 phenotype for suppression. To facilitate efficient co-delivery CA4 PIC enhanced anti-angiogenic immunotherapy, develop injectable metal-organic framework hydrogel using Zeolitic Imidazolate Framework-8 (ZIF-8) hyaluronic acid (HA) (ZIF-8/HA). Our findings demonstrate that ZIF-8 enables loading enhances stability against RNAase degradation

Abstract Bladder cancer (BCa) is a prevalent malignancy with poor prognosis. SLC7A7 has been linked to BCa progression and angiogenesis, but its specific role remains unclear. We established SLC7A7-knockdown cell line investigate effects on angiogenesis. In vivo experiments assessed tumor vascularization, while in vitro studies explored exosome involvement. MiRNA sequencing identified miR-152-3p as key regulator. Further investigation using dual-luciferase reporter assays, qRT-PCR, Western blot revealed that inhibits the expression of FGFR3 by binding 3’ UTR. Meanwhile, functional including angiogenesis Transwell wound healing were performed evaluate confirmed significant progression, specifically promoting through involvement exosomes regulatory axis miR-152-3p/ FGFR3. Abstract The infected chronic wound area has a unique microenvironment featuring hypoxia, excessive reactive oxygen species (ROS), and inflammatory conditions. Bacteria‐formed biofilms limit the anti‐inflammation efficiency of most therapeutics by hindering their penetration into deep tissues increasing drug resistance. unitary modulation pro‐inflammatory M1 macrophage polarization cannot relieve inflammation immediately. Here, “all in one” folic acid‐modified small organic molecule‐based nanoparticles (2TT‐ m C6B@CeO 2 @FA, PCFs) are developed, which possess dual‐targeting to bacteria macrophages, double photothermal therapy (PTT), enzymatic‐like activities. Based on considerable catalase‐ superoxide dismutase‐like activities, PCFs can efficiently scavenge ROS produce (O ). generated O automatically drive movement as nanomotors further promote rescue hypoxia. scavenging promotes macrophages polarized anti‐inflammatory M2 macrophages. study also identifies that FA‐modified target selectively eliminate through PTT, relieves healing. Transcriptome analysis confirms inhibit expression inflammatory‐related genes while cytokines. In vivo experiments identify benefit neovascularization neonatal tissue generation at sites.

Angiogenesis significantly influences the carcinogenesis of thymic epithelial tumors (TET). Both thymomas and carcinoma (TC) overexpress VEGF-A VEGFR-1 -2. This review aims to provide an appraisal use anti-angiogenics in treatment TET. The literature research identified 16 studies that were deemed eligible for further analysis. Seven assessed clinical efficacy sunitinib five apatinib and/or anlotinib. multicenter Japanese phase II REMORA trial investigated lenvatinib, which is a multi-targeted inhibitor VEGFR, FGFR, RET, c-Kit, other kinases. objective response rate was 38% (25.6-52%), highest documented TET progressed after first-line chemotherapy. Anti-angiogenic agents may be useful TET, are not amenable curative treatment. Their toxicity profile seems acceptable. However, angiogenesis inhibitors do appear have major influence on either or TC, although multikinase some effect TC. current evidence suggests most active agent whereas could proposed as acceptable second-line therapy Further concerning combination immune checkpoint with anti-angiogenic drugs warranted.
