Ulcerative colitis (UC) is a debilitating inflammatory bowel disease characterized by intestinal inflammation, barrier dysfunction, and dysbiosis, with limited treatment options available. This study systematically investigates the therapeutic potential of synbiotic composed galactooligosaccharides (GOS) Limosilactobacillus reuteri in murine model colitis, revealing that GOS L. synergistically protect against inflammation dysfunction promoting synthesis pentadecanoic acid, an odd-chain fatty from Bacteroides acidifaciens. Notably, synbiotic, B. acidifaciens, acid are each capable suppressing enhancing tight junction inhibiting NF-κB activation. Dietary polyunsaturated fatty acids (PUFAs), especially ω-3 ω-6, are key regulators responses help maintain integrity barrier. Salmonella enterica serovar Enteritidis (S. Background/Objectives: Dietary fats have been linked to the increasing incidence of chronic diseases, including inflammatory bowel diseases (IBD), namely, Crohn’s disease (CD). Methods: This study investigated impact pentadecanoic acid (C15:0), a type an odd-numbered chain saturated fatty acid, for its potential anti-inflammatory properties in different mouse models experimental IBD using SAMP1/YitFc (SAMP) line (14- or 24-week-old), ileitis and DSS-induced colitis.
