This paper explored the novel approach of targeting cyclic guanosine monophosphate (GMP)-adenosine (AMP) synthase-stimulator interferon genes (cGAS-STING) pathway for treatment osteosarcoma (OS). Osteosarcoma is a common malignancy in adolescents. Most patients die from lung metastasis. It reviewed epidemiology and pathological characteristics OS, highlighting its highly malignant nature tendency pulmonary metastasis, underscoring importance identifying new therapeutic targets. The cGAS-STING was closely associated with biological behaviors OS cells, suggesting that this could be promising strategy. Currently, research on role has been limited, underlying mechanisms remain unclear. Therefore, further investigation into exploration strategies based are great significance developing more effective treatments OS. offered fresh perspective providing hope options by pathway.

Osteosarcoma, an aggressive bone malignancy predominantly affecting children and adolescents, presents significant therapeutic challenges with a 5-year survival rate below 30% in metastatic cases. T-cell exhaustion, characterized by the overexpression of immune checkpoint molecules, contributes to osteosarcoma progression evasion. Although targeting these inhibitory pathways has shown potential restoring activity, molecular regulators depletion are poorly understood. This study employed comprehensive bioinformatics analyses on samples from TARGET database, combined normal tissue data GTEx identify exhaustion-associated genes their co-expressed long non-coding RNAs (lncRNAs). Gene ontology KEGG pathway were used elucidate immune-related enrichments. A six-lncRNA prognostic model was established using LASSO regression validated separate cohorts. Functional assays evaluated impact lncRNA AL031775.1 cell behavior function. Twenty-four key exhaustion-related identified significantly enriched pathways, indicating importance microenvironment. The constructed stratified patients prognosis, showing robust predictive performance across Among six lncRNAs, is notably downregulated promotes proliferation, migration, invasion while contributing exhaustion. In T cells, downregulation impairs antitumor immunity, upregulates molecules LAG3, PD1, CTLA4, diminishes cytotoxic activity against tumor cells. identifies novel highlights managing enhancing immunity counteracting progression. Osteosarcoma (OS) is the prevalent primary bone cancer, with a high proclivity for local invasion and metastasis. Previous studies have indicated that telomeres are closely related to prognosis of but significance telomere-related features in OS remains uncertain. Thus, goal this work identified subtypes based on genes (TRGs). The data was collected from TARGET Gene Expression Omnibus databases. Firstly, we mediated by TRGs OS. Subsequently, analyzed immune characteristics telomeres-related Moreover, built signature via univariate LASSO Cox regression analyses, correlation TME. Dysregulation of R-loops has been implicated in tumor development, progression, and the regulation immune microenvironment (TME). However, their roles osteosarcoma (OS) remain underexplored. In this study, we firstly constructed a novel R-loop Gene Prognostic Score Model (RGPSM) based on RNA-sequencing (RNA-seq) datasets evaluated relationships between RGPSM scores TME. Additionally, identified key R-loop-related genes involved OS progression using single-cell RNA sequencing (scRNA-seq) dataset, validated these findings through experiments. We found that patients with high-RGPSM exhibited poorer prognosis, lower Huvos grades more suppressive Moreover, proportion malignant cells was significantly higher group. And integrated analysis RNA-seq scRNA-seq revealed PC4 SRSF1 Interacting Protein 1 (PSIP1) highly expressed osteoblastic proliferative cells. Notably, high expression PSIP1 associated poor prognosis patients. Subsequent experiments demonstrated knockdown inhibited both vivo vitro, leading increased accumulation DNA damage. Conversely, overexpression facilitated resolution reduced damage induced by cisplatin. conclusion, developed effectively predicted outcomes across diverse cohorts as critical modulator regulating
