Abstract Background Prostate cancer (PCa) is a leading malignancy with rising global incidence, posing significant challenges in treatment. The immunosuppressive tumor microenvironment (TME) CRPC major barrier to effective immunotherapy. Identifying therapeutic targets that modulate the immune response within TME crucial for advancing PCa Materials and Methods In this study, we employed Mendelian randomization (MR) investigate causal relationships between gene expression of blood proteins risk. We utilized cis-eQTL data from eQTLGen Consortium cell phenotype NHGRI-EBI GWAS Catalog. Our analysis included discovery validation cohorts, totaling over 800,000 individuals. Colocalization was performed confirm genetic associations, mediation MR used explore mediating role immunity. Background Colorectal cancer (CRC) is the second leading cause of deaths globally, which poses a heavy burden on our healthcare and economy. In recent years, increasing researches suggest that tumor microenvironment (TME) influences onset, progression, metastasis, treatment. This has become popular direction for researching attacking cancer. However, to date, there no bibliometric analysis colorectal from 2014 2024. study aims provide comprehensive picture current research status, hotspots, future trends in this field perspective. Methods study, publications about 2024 were searched based Web Science Core Collection database. Then we analyzed visualized data using CiteSpace, VOSviewer, bibliometrix package, Microsoft Excel 2019. Colorectal cancer (CRC) stands as one of the tumors with globally high incidence and mortality rates. In recent years, researchers have extensively explored role tumor immune microenvironment (TME) in CRC, highlighting crucial influence cell populations driving progression shaping therapeutic outcomes.
